针刺调控氧化应激对肥胖雄鼠生殖损伤的保护作用及机制研究OA
Acupuncture ameliorates oxidative stress-related reproductive damage in obese male mice
目的:探讨针刺通过调节氧化应激改善肥胖雄性小鼠生殖损伤的可能机制.方法:小鼠随机分为空白组(n=18)和高脂饮食组(n=35).空白组正常饲料喂养,高脂饮食组给予高脂饲料喂养,10周后,称重筛选肥胖小鼠.取造模成功的肥胖小鼠,随机分为模型组和针刺组(n=15).针刺组小鼠给予针刺干预,连续4周.检测精子质量;ELISA法检测血清总胆固醇(TC)、甘油三酯(TG)、高密度脂蛋白胆固醇(HDL-C)、低密度脂蛋白胆固醇(LDL-C)、睾酮(T)、促卵泡生成素(FSH)、促黄体生成激素(LH)含量;HE和油红O染色观察睾丸病理形态;生化法检测睾丸丙二醛(MDA)、还原型谷胱甘肽(GSH)和总超氧化物歧化酶(T-SOD)含量;流式细胞术检测睾丸活性氧(ROS)水平;透射电镜观察睾丸线粒体结构;TUNEL染色法检测睾丸阳性细胞凋亡率;Western blot法检测睾丸肿瘤蛋白p53(p53)、B淋巴细胞瘤2(BCL-2)、B淋巴细胞瘤2关联X的蛋白(BAX)、半胱氨酸蛋白酶3(Caspase-3)蛋白表达.结果:与空白组比较,模型组精子浓度和活力降低(P<0.01,P<0.05);血清TC、TG、LDL-C含量提高(P<0.01),HDL-C含量降低(P<0.01);血清T、FSH和LH含量降低(P<0.01);睾丸内生精小管结构损伤,出现较多脂质沉积;MDA含量升高(P<0.01),GSH和T-SOD含量降低(P<0.01);ROS水平升高(P<0.01);线粒体形态变差;阳性细胞凋亡率升高(P<0.01);BCL-2蛋白表达降低(P<0.01),BAX、Caspase-3、p53蛋白表达升高(P<0.01).与模型组比较,针刺组精子浓度和活力升高(P<0.01,P<0.05);血清HDL-C含量升高(P<0.01),TC、TG、LDL-C含量降低(P<0.01,P<0.05);T、FSH和LH含量升高(P<0.01,P<0.05);睾丸病理形态和脂质沉积改善;MDA含量降低(P<0.01),GSH和T-SOD含量升高(P<0.01,P<0.05);ROS水平降低(P<0.01);线粒体数量和形态改善;阳性细胞凋亡率降低(P<0.01);BCL-2蛋白表达升高(P<0.05),BAX、Caspase-3、p53蛋白表达降低(P<0.01,P<0.05).结论:针刺可以调节氧化应激水平,改善线粒体功能,抑制生精细胞过度凋亡,从而改善肥胖相关的雄性小鼠生殖损伤.
Objective:To investigate whether acupuncture ameliorates reproductive injury in obese male mice by regulating oxidative stress.Methods:Mice were randomly divided into a control group(n=18)and a high-fat diet group(n=35).The control group was fed a normal diet,while the high-fat diet group was fed a high-fat diet.After 10 weeks,body weight was measured and obese mice were screened.Successfully established obese mice were ran-domly divided into a model group and an acupuncture group(n=15).The acupuncture group received acupuncture intervention for 4 consecutive weeks.Sperm quality was evaluated.Serum levels of total cholesterol(TC),triglycer-ides(TG),high-density lipoprotein cholesterol(HDL-C),low-density lipoprotein cholesterol(LDL-C),testosterone(T),follicle-stimulating hormone(FSH),and luteinizing hormone(LH)were detected by ELISA.Testicular patho-logical morphology was observed by hematoxylin-eosin(HE)staining and Oil Red O staining.The contents of ma-londialdehyde(MDA),reduced glutathione(GSH),and total superoxide dismutase(T-SOD)in testicular tissue were measured by biochemical methods.Reactive oxygen species(ROS)levels in the testes were detected by flow cytome-try.Testicular mitochondrial structure was observed by transmission electron microscopy.The apoptosis rate of posi-tive cells in the testes was detected by TUNEL staining.Protein expression levels of tumor protein p53(p53),B-cell lymphoma-2(BCL-2),BCL-2-associated X protein(BAX),and cysteine protease-3(Caspase-3)were detected by Western blot.Results:Compared with the control group,the model group showed decreased sperm concentration and motility(P<0.01,P<0.05);increased serum TC,TG,and LDL-C levels(P<0.01)and decreased HDL-C levels(P<0.01);decreased serum T,FSH,and LH levels(P<0.01);structural damage of seminiferous tubules in the testes with increased lipid deposition;increased MDA content(P<0.01)and decreased GSH and T-SOD contents(P<0.01);elevated ROS levels(P<0.01);impaired mitochondrial morphology;increased apoptosis rate of positive cells(P<0.01);decreased BCL-2 protein expression(P<0.01);and increased expression of BAX,Caspase-3,and p53 proteins(P<0.01).Compared with the model group,the acupuncture group showed increased sperm concentra-tion and motility(P<0.01,P<0.05);increased serum HDL-C levels(P<0.01)and decreased TC,TG,and LDL-C levels(P<0.01,P<0.05);increased T,FSH,and LH levels(P<0.01,P<0.05);improved testicular pathological morphology and reduced lipid deposition;decreased MDA content(P<0.01)and increased GSH and T-SOD con-tents(P<0.01,P<0.05);reduced ROS levels(P<0.01);improved mitochondrial number and morphology;de-creased apoptosis rate of positive cells(P<0.01);increased BCL-2 protein expression(P<0.05);and decreased BAX,Caspase-3,and p53 protein expression(P<0.01,P<0.05).Conclusion:Acupuncture can regulate oxidative stress levels,improve mitochondrial function,and inhibit excessive apoptosis of spermatogenic cells,thereby impro-ving obesity-related sperm quality decline.
