超临界CO2萃取侧柏叶提取物对雄激素性脱发的防治功效及机制研究OA
Study on the efficacy and mechanism of Platycladus orientalis extracted by supercritical CO2 in prevention and treatment of androgenetic alopecia
本研究旨在探究超临界CO2萃取侧柏叶提取物(SC-CO2-PO)的防脱发作用机制.采用超临界CO2萃取技术制备侧柏叶提取物,通过气相色谱-质谱联用技术(GC-MS)进行全成分解析及特征活性成分定量分析,建立基于AI的靶点预测模型筛选防脱关键靶点,采用MTT法测定SC-CO2-PO对人皮脂腺细胞(SZ95)的毒性作用,采用尼罗红荧光染色评价其对中性脂质抑制效果,采用酶活实验检测对II型5α-还原酶(SRD5A2)的抑制率,采用半定量RT-PCR检测雄激素受体(AR)mRNA相对表达水平,通过离体毛囊模型观察毛囊生长周期调控效应.结果表明,SC-CO2-PO含25种活性成分,其特征性活性成分雪松醇和β-石竹烯质量分数分别1.32%和0.10%;SC-CO2-PO在SZ95中的安全质量浓度为0.019 5 mg/mL;SC-CO2-PO能抑制中性脂质合成,抑制SRD5A2活性,减少AR mRNA相对表达含量,上调血管内皮生长因子(VEGF)表达水平和增加毛囊长度.这些结果表明,SC-CO2-PO能通过双途径起到防脱发作用.SC-CO2-PO通过抑制SRD5A2活性和下调AR mRNA相对表达水平,调控雄激素信号通路来减少皮脂分泌;SC-CO2-PO上调VEGF表达水平,在毛囊周期中促使毛囊周围血管重建,提高毛囊活性,促进头发的生长,延长毛囊生长期.
The aim of this study is to investigate the anti-hair loss mechanism of supercritical CO2 extraction of Platycladus orientalis(L.)Franco(SC-CO2-PO).Supercritical CO2 extraction technology was used to prepare the extract of Platycladus orientalis(L.)Franco.Full component analysis and quantification of the characteristic active ingredients were performed by gas chromatography-mass spectrometry(GC-MS).An AI-based target prediction model was established to screen key anti-shedding targets.The cytotoxic effect of SC-CO2-PO on human sebaceous gland cells(SZ95)was determined by MTT assay.Nile red fluorescence staining was used to evaluate its inhibitory effect on lipid synthesis.Enzyme activity assays were used to measure the inhibition rate of type II 5α-reductase(SRD5A2).Semi-quantitative RT-PCR was used to detect the relative expression levels of androgen receptor(AR)mRNA.An in vitro hair follicle culture model was used to observe its regulatory effect on the hair follicle growth cycle.The results show that SC-CO2-PO contains 25 active ingredients,with the contents of the characteristic active ingredients cedarol and β-caryophyllene being 1.32%and 0.10%,respectively.The safe concentration of SC-CO2-PO in SZ95 cells is 0.019 5 mg/mL.SC-CO2-PO can inhibit neutral lipid synthesis,suppress SRD5A2 activity,reduce the relative expression levels of AR mRNA,increase hair follicle length and vascular endothelial growth factor(VEGF)content.These results indicate that SC-CO2-PO exerts anti-hair loss effects via dual pathways:it reduces sebum secretion by inhibiting SRD5A2 activity and down-regulating AR mRNA expression,thereby modulating the androgen signaling pathway.SC-CO2-PO increases the expression level of VEGF,promoting the reconstruction of blood vessels around hair follicles during the hair follicle cycle,enhancing the activity of hair follicles,stimulating hair growth,and prolonging the growth period of hair follicles.
