四川省乐山市主栽晾晒烟品种烟叶品质特征和代谢物差异分析OA
Analysis of Quality Characteristics and Metabolite Differences of Flue-Cured Tobacco Leaves of Main Sun-Cured Tobacco Varieties in Leshan City,Sichuan Province
为明确四川省乐山市主栽晾晒烟品种烟叶的品质特征和代谢物差异,以聂尔青青毛(NQ)和聂尔青黄毛(NH)为供试品种,晾制发酵后,采用固相微萃取(SPME)结合气相色谱-质谱联用技术(GC-MS),对其烟叶外观品质、物理性状、化学成分和感官品质进行分析,并检测香气成分和代谢物.结果表明,从外观品质看,NQ烟叶外观品质优于NH,更符合优质晾晒烟的评价标准.从物理性状看,NH烟叶的叶质质量、平衡含水率和抗张强度均显著高于NQ,最大叶长、最大叶宽、单叶质量、叶厚和含梗率与NQ均无显著差异.从化学成分看,NH烟叶的总糖含量、还原糖含量、烟碱含量、氯含量和钾含量均显著高于NQ,钾氯比显著低于NQ.从感官品质看,NH烟叶的香气层次、舒适度与感官协调性均优于NQ.从香气成分看,NH烟叶7种香气物质含量显著高于NQ,分别为6,10,14-三甲基-2-十五烷酮、1-甲基-2-吡咯烷酮、可替宁、(1'S,2'S)-尼古丁-N'-氧化物、六甲基环三硅氧烷、1,2,3,6-四氢-2,3'-联吡啶和1-(3-吡啶基)乙酮.从代谢物差异看,NH烟叶8种代谢物含量显著高于NQ,分别为鸟苷-5'-一磷酸、2-脱氧尿苷-5'-单磷酸、L-胱硫醚、γ-L-谷氨酰-L-2-氨基丁酸酯、异柠檬酸、L-酪氨酸、L-谷氨酰胺和5-脱氧-5-甲硫腺苷,1种代谢物含量显著低于NQ,为胞嘧啶.此外,KEGG富集结果表明,NQ和NH烟叶差异代谢物显著富集于20条通路,与植物氮素代谢、能量代谢及次生代谢物合成密切相关.研究结果为乐山市优质晾晒烟品种的选用提供了理论基础.
In order to clarify the quality characteristics and metabolite differences of the main air-cured tobacco varieties in Leshan County,Sichuan Province,Nie'er QING Qingmao(NQ)and Nie'er Qing Huangmao(NH)were used as test materials.After air-curing and fermentation,solid phase microextraction(SPME)combined with gas chromatography-mass spectrometry(GC-MS)was used to analyze totacco leaves appearance quality,physical properties,chemical composition and sensory quality,and to detect totacco leaves aroma components and metabolites.The results showed that the appearance quality of NQ tobacco leaves was better than that of NH,which was more in line with the evaluation criteria of high-quality air-cured tobacco.From the perspective of physical properties,the leaf weight,equilibrium moisture content and tensile strength of NH tobacco leaves were significantly higher than those of NQ,and the maximum leaf length,maximum leaf width,single leaf weight,leaf thickness and stem content were not significantly different from those of NQ.From the perspective of chemical composition,the total sugar content,reduced sugar content,nicotine content,chlorine content and potassium content of NH tobacco leaves were significantly higher than those of NQ,and the ratio of potassium to chlorine was significantly lower than that of NQ.From the perspective of sensory quality,the aroma level,comfort and sensory coordination of NH tobacco leaves were due to NQ.From the perspective of aroma components,the content of seven aroma substances in NH tobacco leaves was significantly higher than that of NQ,which were 6,10,14-trimethyl-2-pentadecanone,1-methyl-2-pyrrolidone,cotinine,(1'S,2'S)-nicotine-N'-oxide,hexamethylcyclotrisiloxane,1,2,3,6-tetrahydro-2,3'-bipyridine and 1-(3-pyridyl)ethanone.From the difference of metabolites,the contents of 8 metabolites in NH tobacco leaves were significantly higher than those in NQ,which were guanosine-5'-monophosphate,2-deoxyuridine-5'-monophosphate,L-cystathionine,γ-L-glutamyl-L-2-aminobutyric acid ester,isocitrate,L-tyrosine,L-glutamine and 5-deoxy-5-methylthioadenosine,respectively.The content of one metabolite was significantly lower than that of NQ,which was cytosine.In addition,KEGG enrichment results showed that the differential metabolites of NQ and NH tobacco leaves were significantly enriched in 20 pathways,which were closely related to plant nitrogen metabolism,energy metabolism and secondary metabolite synthesis.The results provide a theoretical basis for the selection of high-quality sun-cured tobacco varieties in Leshan City.
