捕食线虫性真菌Duddingtonia flagrans DF-shz3株的分离鉴定及其捕食活性与产孢特性研究OA
Isolation and Identification of The Nematode-Trapping Fungus Duddingtonia flagrans Strain DF-shz3 and Its Predatory Activity and Sporulation Characteristics
研究旨在分离出具有高捕食线虫活性和产孢能力的捕食线虫性真菌,为草食动物线虫病的生物防治制剂的研发提供候选菌种.对新疆天山牧场中采集的土壤样品进行捕食性真菌的分离鉴定,根据分离株的形态学特征与分子生物学结果进行种属鉴定,并对其捕食活性与产孢特性进行测定.成功分离出1株捕食线虫性真菌(命名为DF-shz3),该菌菌落呈白色,菌丝分隔分枝,可产生大量的厚垣孢子和少量的分生孢子;厚垣孢子呈球状,表面有球形突起;分生孢子呈梨形或椭圆形,有一个分隔,位于孢子中央.遗传进化分析显示,DF-shz3株ITS序列与Duddingtonia flagrans的同源性为100%,与KP257593.1位于同一分支.形态学与分子生物学鉴定结果均表明,DF-shz3分离株为Duddingtonia flagrans.DF-shz3对秀丽隐杆线虫三期幼虫的捕食率为97.93%,捕食结构为黏性菌网.在YPSSA培养基上30 ℃培养28 d,Duddingtonia flagrans DF-shz3可产生数量最多的厚垣孢子.本试验分离出了一株适合新疆本土草食动物线虫病防控的捕食线虫真菌Duddingtonia flagrans.
This study aimed to isolate a nematode-trapping fungal strain with high predatory activity and strong sporulation capacity,providing a candidate microorganism for the development of biocontrol a-gents against nematode infections in herbivorous animals.Soil samples were collected from the Tianshan Pasture in Xinjiang,China,and screened for nematode-trapping fungi.The isolated strains were identified based on morphological characteristics and molecular phylogenetic analysis.Their predatory efficiency and sporulation traits were also evaluated.A single strain,designated DF-shz3,was successfully isolated.The colony appeared white,with septate and branched hyphae,and produced abundant chlamydospores and a small number of conidia.The chlamydospores were globose with spherical surface ornamentations,while the conidia were pyriform or ovoid with a single septum located centrally.Phylogenetic analysis based on the ITS sequence revealed 100%homology between strain DF-shz3 and Duddingtonia flagrans,clustering with the reference sequence KP257593.1.Both morphological and molecular evidence confirmed that DF-shz3 belongs to Duddingtonia flagrans.Strain DF-shz3 exhibited a high predation rate of 97.93%against third-stage larvae of Caenorhabditis elegans,forming adhesive networks as its trapping structure.When cultured on YPSSA medium at 30℃ for 28 days,DF-shz3 produced the highest yield of chlamydospores.In this study,a strain of the nematode-trapping fungus Duddingtonia flagrans suitable for the prevention and control of nematodiasis in native herbivorous animals in Xinjiang was isolated.
