Effects of Slow-released Urea on Rumen Tissue Morphology,Proteinase Activity and Microbial Diversity of Tibetan Cashmere Goats

试验旨在研究缓释包被尿素对藏西北绒山羊瘤胃组织形态、蛋白酶活性及微生物多样性的影响.选取15月龄藏西北绒山羊30只(公母各半),按单因素水平分为3组,每组10个重复,每个重复1只羊.分为对照组、尿素组(2%)和缓释尿素组(2.4%),预试期7 d,正试期49 d.结果表明:(1)尿素组瘤胃乳头高度和角质厚度低于对照组和缓释尿素组(P<0.05);(2)缓释尿素组瘤胃蛋白酶活力和胃蛋白酶活性高于对照组和尿素组(P<0.05);(3)与对照组和缓释尿素组相比,尿素组放线菌门(Actinobacteriota)、绿弯菌门(Chloroflexi)(P<0.01)和厚壁菌门(Firmicutes)(P<0.05)相对丰度升高,而拟杆菌门(Bacteroidota)相对丰度降低(P<0.05);缓释尿素组髌骨细菌门(Patescibacteria)相对丰度高于对照组和尿素组(P<0.01),且对照组疣微菌门(Verrucomicrobiota)相对丰度高于尿素组和缓释尿素组(P<0.05).与对照组和缓释尿素组相比,尿素组克里斯滕森菌科R-7群(Christensenellaceae_R-7_group)(P<0.01)、未分类的厚壁菌(Firmicutes_unclassified)和 NK4A214 群(NK4A214_group)(P<0.05)相对丰度升高,理研菌科 RC9 肠道群(Rikenellaceae_RC9_gut_group)(P<0.01)和普氏菌属(Prevotella)(P<0.05)相对丰度降低;尿素组细菌F082相对丰度低于对照组(P<0.05);缓释尿素组候选单胞生糖菌属(Candidatus_Saccharimonas)相对丰度高于对照组和尿素组(P<0.01).综上,饲粮添加尿素改变了绒山羊的瘤胃菌群组成;相比于尿素,饲粮添加缓释尿素能够提高瘤胃蛋白酶活性,且对绒山羊瘤胃发育不产生负面影响.

To investigate the effects of slow-released urea on rumen morphology,proteinase activity and microbial diversity of Tibetan cashmere goats,thirty 15-month-old Tibetan cashmere goats(half male and half female)were divided into 3 groups with 10 replicates per group and 1 sheep per replicate according to single factor level.They were divided into control group,urea group(2%)and slow-released urea group(2.4%).The pre-test period lasted for 7 days and the trial period lasted for 49 days.The results showed as follows:(1)the rumen papillary height and keratin thickness in urea group were lower than those in con-trol group and slow-released urea group(P<0.05).(2)The rumen protease activity and pepsin activity in slow-released urea group were higher than those in control group and urea group(P<0.05).(3)Com-pared with control group and slow-released urea group,the relative abundance of Actinobacteriota,Chlo-roflexi(P<0.01)and Firmicutes(P<0.05)in urea group was increased and Bacteroidota was decreased(P<0.05).The rumen of cashmere goats in the slow-released urea group had higher relative abundance of Patescibacteria than that in the control group and the urea group(P<0.01),and the rumen of cashmere goats in the control group had higher relative abundance of Verrucomicrobiota than that in the urea group and the slow-released urea group(P<0.05).Compared with the control group and the slow-released urea group,the relative abundance of Christensenellaceae_R-7_group in urea group(P<0.01),Firmicutes_un-classified and NK4A214 group(NK4A214_group)were increased(P<0.05),while Rikenellaceae_RC9_gut_group(P<0.01)and Prevotella(P<0.05)were decreased.The rumen of cashmere goats in the urea group had lower relative abundance of bacteria F082 than that in the control group(P<0.05).The relative abundance of Candidatus_Saccharimonas in slow-released urea group was higher than that in control group and urea group(P<0.01).In conclusion,dietary urea supplementation changed rumen microflora structure of cashmere goats.Compared with urea,dietary supplemented with slow-released urea could in-crease rumen protease activity,and has no negative effect on rumen development of cashmere goats.
