不同品系德州驴盲肠微生物组成的比较研究OA
Comparative Study on Microbial Composition of Cecum of Different Strains in Dezhou Donkeys
试验旨在研究不同品系德州驴盲肠微生物组成的差异.随机选取11月龄、体重(185±3)kg的健康公驴20头,分为三粉驴组(n=10)和乌头驴组(n=10),每组各10头驴.2个组均饲喂饲粮组成和营养水平相同的颗粒状全混合饲粮(TMR),试验期100 d,预试期14 d,正试期86 d.饲养试验结束后屠宰试验驴,取盲肠的内容物用于测定盲肠的微生物组成.结果表明:(1)共鉴定出29个门、51个纲、114个目、180个科、265个属.(2)三粉驴和乌头驴盲肠微生物Chao、ACE、Shannon、Simpson等Alpha多样性指数均无显著差异(P>0.05),即菌群丰富度与多样性无显著差异;PCoA图显示三粉驴和乌头驴菌群仅有部分重合,但无法明显分开.(3)在门水平上,三粉驴和乌头驴均以厚壁菌门(Firmicutes)和拟杆菌门(Bacteroid-ota)为主,相对丰度均大于75%;在属水平上,三粉驴在属水平上排名前五的菌属从大到小依次为考拉杆菌属(Phascolarctobacterium)、瘤胃菌科 UCG-005 菌属(Ruminococcaceae_UCG-005),Alloprevotella、链球菌属(Strep tococcus)和嗜黏蛋白阿克曼菌属(Akkermansia);乌头驴在属水平上排名前五的菌群从大到小分别为克里斯滕森氏菌科R-7群(Christensenellaceae_R-7_group)、甲烷短杆菌属(Methanobrevibacter)、链球菌属(Streptococcus)、理研菌科_RC9肠群(Rikenellaceae_RC9_gut_group)和嗜黏蛋白阿克曼菌属(Akkermansia);乌头驴的F/B值(厚壁菌门和拟杆菌门比值)在数值上明显高于三粉驴,但未达到显著性差异(P>0.05).(4)在门水平上,有3种差异性微生物;在属水平上,有9种差异性微生物.综上所述,德州驴盲肠内的微生物复杂多样;三粉驴和乌头驴盲肠微生物丰富度和多样性无差异,盲肠菌群结构差异不大.
This study aims to investigate the differences in the microbial composition of the cecum a-mong different breeds of Dezhou donkeys.A total of 20 healthy male donkeys,aged 11 months and weig-hing(185±3)kg,were randomly selected and divided into two groups:the Sanfen donkey group(n=10)and the Wutou donkey group(n=10),10 donkeys in each group.Both groups were fed a pelleted total mixed ration(TMR)with the same formulation and nutritional level for a pre-feeding period of 14 days,followed by a formal feeding period of 86 days,totaling 100 days.After the feeding trial,the donkeys were slaughtered,and the contents of the cecum were collected to analyze the microbial composition.Results showed that(1)a total of 29 phyla,51 classes,114 orders,180 families,and 265 genera were identified.(2)The alpha diversity indices-including Chao,ACE,Shannon,and Simpson-showed no significant differ-ence between the Sanfen and Wutou donkeys(P>0.05),indicating that the microbial richness and diversi-ty were similar.The Principal Coordinates Analysis(PCoA)plot demonstrated partial overlap between the two groups,but no clear separation was observed.(3)At the phylum level,both breeds predominantly harbored Firmicutes and Bacteroidota,accounting for more than 75%relative abundance.At the genus level,the top five genera in the Sanfen donkey,ranked from high to low,were Phascolarctobacterium,Ruminococcaceae_UCG-005,Alloprevotella,Streptococcus,and Akkermansia;in the Wutou donkey,the top five genera were Christensenellaceae_R-7_group,Methanobrevibacter,Streptococcus,Rikenellaceae_RC9_gut_group,and Akkermansia.The F/B ratio(the ratio of Firmicutes to Bacteroidota)was numeri-cally higher in Wutou donkeys but did not reach significant differences(P>0.05).(4)At the phylum lev-el,there were three differentially expressed microorganisms,and at the genus level,there were nine differentially expressed microorganisms.In summary,the microbial community in the cecum of Dezhou donkeys is complex and diverse;there were no significant differences in microbial richness and diversity between the two breeds,and the PCoA analysis revealed that their microbial community structures are largely similar,with overall minimal differences.
