坝上长尾鸡腿肌脂肪酸含量相关基因多态性及其关联分析OA
Polymorphisms of Genes Related to Fatty Acid Content and Their Association Analysis in BaShang Long-Tailed Chickens
研究探讨了坝上长尾鸡腿肌脂肪酸含量与基因型之间的关联.试验样本分为高脂组和低脂组(各20只),对61只鸡进行全基因组测序,测序深度达10倍以上.利用FST统计量分析高低脂组的遗传分化,筛选出显著选择信号的基因区域,并通过功能注释及GO和KEGG富集分析鉴定出与脂肪合成和代谢相关的候选基因.结果表明,高脂组的平均脂肪酸含量为(8.866±2.52)g/100 g,低脂组为(2.3885±0.45)g/100g.基于FST的滑动窗口分析,共识别出16333个正选择区域,涉及889个基因.显著差异基因富集于脂肪细胞分化通路;BCAT1、PDGFRA、MAP3K5、ANGPT1、SCD和ACOX3基因与脂肪酸代谢显著相关.MAP3K5、BCAT1、PDGFRA 和ANGPT1基因的外显子与脂肪酸含量显著关联(P<0.05).本研究识别的候选基因及其错义突变位点揭示了坝上长尾鸡脂肪沉积的遗传机制,为家禽脂肪性状的遗传改良提供了重要的分子标记.
This study investigated the association between fatty acid content in the thigh muscle of Bas-hang Long-tailed chickens and their genotypes.The experimental samples were divided into high-fat and low-fat groups(each consisting of 20 individuals).Whole-genome sequencing was performed on 61 chick-ens,with sequencing depths exceeding 10 ×.Genetic differentiation between the high-and low-fat groups was analyzed using FST statistics to identify genomic regions under selection.Candidate genes related to fat synthesis and metabolism were identified through functional annotation,Gene Ontology(GO),and KEGG pathway enrichment analyses.The results showed that the average fatty acid content in the high-fat group was 8.866±2.52 g/100 g,while in the low-fat group it was 2.3885±0.45 g/100 g.Based on a sliding win-dow analysis of FST,a total of 16,333 selective sweep regions were identified,involving 889 genes.GO en-richment analysis indicated that the differentially expressed genes were significantly overrepresented in pathways related to adipocyte differentiation;KEGG pathway analysis revealed that genes such as BCAT1,PDGFRA,MAP3K5,ANGPT1,SCD,and ACOX3 were significantly associated with fatty acid metabolism.PCR sequencing validation demonstrated that exonic regions of MAP3K5,BCAT1,PDGFRA,and ANGPT1 were significantly correlated with fatty acid content(P<0.05).The candidate genes and their non-synonymous mutation sites identified in this study elucidate the genetic mechanisms underlying fat deposition in Bashang Long-tailed chickens and provide valuable molecular markers for the genetic improvement of fat-related traits in poultry.
