水产动物抗菌肽重组表达与作用机制研究进展OA
Research progress on recombinant expression and mechanisms of action of antimicrobial peptides in aquatic animals
随着水产养殖集约化程度的不断提高,水产动物细菌性、病毒性和真菌性疾病频发,传统抗生素的长期使用引发耐药性、药物残留及生态风险等问题,促使新型抗感染替代物的研究受到关注.抗菌肽是水产动物先天免疫系统的重要效应分子,具有广谱抑菌、作用机制多样及耐药性产生风险较低等特点.近年来,鱼类、甲壳类和软体动物等水产动物抗菌肽的结构特征、生物学功能及进化多样性研究不断深入,其在疾病防控和免疫调控中的作用逐渐得到阐明.同时,随着分子生物学和生物工程技术的发展,抗菌肽的重组表达体系不断完善,为其规模化制备和工程化应用提供了技术基础.本文综述了水产动物抗菌肽的主要类型及其生物学功能,总结了不同异源表达体系中抗菌肽重组表达的研究进展,并对其直接抗菌、免疫调控以及抗病毒和抗真菌作用机制进行了概述,以期为抗菌肽在水产养殖中的进一步研究与应用提供参考.
With the increasing intensification of aquaculture,bacterial,viral,and fungal diseases in aquatic animals have become increasingly prevalent.The long-term use of traditional antibiotics has led to problems such as antimicrobial resistance,drug residues,and ecological risks,prompting growing interest in the development of novel anti-infective alternatives.Antimicrobial peptides(AMPs)are important effector molecules of the innate im-mune system of aquatic animals and are characterized by broad-spectrum antimicrobial activity,diverse mechanisms of action,and a relatively low risk of inducing drug resistance.In recent years,extensive studies have been conducted on the structural characteristics,biological functions,and evolutionary diversity of antimicrobial peptides derived from aquatic animals,including fish,crustaceans,and mollusks,and their roles in disease pre-vention and immune regulation have been progressively elucidated.Meanwhile,advances in molecular biology and bioengineering have led to continuous improvements in recombinant expression systems for antimicrobial peptides,providing a technical basis for large-scale production and practical applications.This review summarizes the major types and biological functions of antimicrobial peptides in aquatic animals,outlines recent progress in their recombi-nant expression using different heterologous expression systems,and discusses their mechanisms of action,including direct antibacterial activity,immunomodulatory effects,as well as antiviral and antifungal activities,with the aim of providing a reference for further research and application of antimicrobial peptides in aquaculture.
