为探究组织蛋白酶H(CTSH)与猪非典型瘟病毒云南株(APPV-YN)Mut蛋白的相互作用及其对APPV复制的影响,首先利用ProtParam等在线软件对CTSH基因表达蛋白的理化性质、亚细胞定位等特征进行分析预测;通过GST pull-down和双分子荧光互补(BiFC)技术分别验证CTSH与APPV-YN Mut的体外互作及细胞内的相互作用;采用qPCR和 Western blot确定过表达或敲低宿主蛋白CTSH对APPV复制的影响.结果显示,CTSH蛋白是亲水性稳定蛋白,二级结构主要以无规则卷曲为主,三级结构稳定,可能主要位于细胞质中;Mut与宿主蛋白CTSH在体外和细胞内都存在相互作用;在细胞中过表达CTSH,APPV的基因组拷贝数与蛋白表达水平显著上升;反之,利用siRNA敲低CTSH后,APPV增殖被明显抑制.综上所述,CTSH与Mut相互作用并促进APPV的复制.该结果不仅有助于深入理解病毒与宿主细胞的相互作用机制,还可能为非典型瘟病等病毒性疾病的防控提供新的思路和方法.

In order to explore the interaction between Cathepsin H(CTSH)and Mut protein of Atypical Porcine pestivirus-Yunnan(APPV-YN)and its effect on APPV replication,the physicochemical properties and subcellular localization of CTSH gene expression protein were analyzed and predicted by online soft-ware such as ProtParam.The interaction between CTSH and APPV-YN Mut in vitro and in vivo was veri-fied by GST pull-down and Bimolecular fluorescence complementation(BiFC),respectively.The effects of overexpression or knockdown of host protein CTSH on APPV replication were determined by qPCR and Western blot.The results showed that CTSH protein was a hydrophilic stable protein,the secondary struc-ture was mainly random coil,and the tertiary structure was stable,which may be mainly located in the cy-toplasm.Mut interacts with the host protein CTSH in vitro and in vivo.Overexpression of CTSH in cells significantly increased the genome copy number and protein expression level of APPV.On the contrary,af-ter knocking down CTSH with siRNA,APPV proliferation showed a significant inhibitory effect.In sum-mary,CTSH interacts with Mut and promotes APPV replication.The results not only help to understand the interaction mechanism between virus and host cells,but also provide new ideas and methods for the prevention and treatment of viral diseases such as atypical plague.
