猫血浆中氟雷拉纳液相色谱-串联质谱检测方法的建立OA
Establishment of an LC-MS/MS Method for Fluralaner Determination in Cats' Plasma
氟雷拉纳(Flur)为异恶唑啉类杀虫、杀螨剂,具有易吸收、安全、高效、长效和良好的耐受性等特点.为研究复方Flur滴剂在猫体内的药代动力学,以Flur-D4为内标,建立了猫血浆中Flur的液相色谱-串联三重四级杆质谱联用技术(LC-MS/MS)检测方法.以乙腈为提取溶剂,水浴超声辅助提取,减压蒸干后复溶,LC-MS/MS检测,内标法定量.结果显示,猫血浆样品基质对Flur的定量检测无干扰,方法特异性良好;方法灵敏度高,检测限(LOD)为5 ng/mL,定量限(LOQ)为10 ng/mL;基质效应不显著,以溶剂标准溶液制备标准曲线,Flur在10~6 000 ng/mL的浓度范围内,线性关系良好(R2>0.99);在不同浓度的添加水平下,平均准确度在92.60%~116.70%之间,批内、批间变异系数均小于10%,符合方法学要求.不同浓度加标样品在-18 ℃以下冷冻保存6个月稳定,或反复冻融后均稳定,不同浓度加标样品的处理液4 ℃放置72 h稳定.标准储备液、标准中间储备液9个月内稳定,标准工作液2个月内稳定.结果表明,建立的方法特异性好,灵敏度高,稳定可靠,可用于Flur滴剂在猫体内药代动力学的研究.
Fluralaner(Flur)is an isoxazoline insecticide and acaricide characterized by rapid absorption,safety,high efficacy,long-lasting effects,and good tolerability.To investigate the pharmacokinetics of com-pound Flur drops in cats,a liquid chromatography—tandem quadrupole mass spectrometry(LC-MS/MS)method for the detection of Flur in feline plasma was established using Flur-D4 as an internal standard.Acetonitrile was used as the extraction solvent,with water-bath ultrasonic-assisted extraction followed by vacuum evaporation and redissolution prior to LC-MS/MS analysis and quantification via the internal standard method.The results showed that the feline plasma matrix did not interfere with the quantitative detection of Flur,and the method exhibited good specificity;The method exhibited high sensitivity,with a limit of detection(LOD)of 5 ng/mL and a limit of quantification(LOQ)of 10 ng/mL;matrix effects were insignificant.A standard curve was prepared using solvent-based standard solutions,and Flur exhibited a good linear relationship(R2>0.99)in the concentration range of 10 ng/mL to 6 000 ng/mL.The spiked samples with different concentration levels,the mean accuracy ranged from 92.60%to 116.70%,with both intra-and inter-batch coefficients of variation below 10%,meeting methodological requirements.Spiked samples with different concentration remained stable when stored frozen at-18℃ or below for 6 months or after repeated freeze-thaw cycles;and the processed solution of spiked samples remained stable when stored at 4℃ for 72 hours.The standard stock solution and intermediate standard solution were stable for 9 months,while the working standard solution was stable for 2 months.The results indicate that the estab-lished method exhibits good specificity,high sensitivity,and is stable and reliable,making it suitable for pharmacokinetic studies of Flur drops in cats.
