郁金散改良组方对鸡大肠埃希氏菌腹泻模型的治疗效果评价OA
Evaluation of Therapeutic Effect of Yujin Powder Modified Formula on Chicken E.coli Diarrhea Model
为明确郁金散改良组方对鸡大肠埃希氏菌腹泻的治疗作用及给药剂量.将14 d芦花鸡分为空白对照组(Ⅰ)、模型对照组(Ⅱ)、郁金散改良组方1/3倍(Ⅲ)、1/2倍(Ⅳ)、1倍(Ⅴ)、2倍(Ⅵ)、3倍(Ⅶ)剂量组和氟苯尼考组(Ⅷ).除CON外,各组雏鸡用抗菌药物处理后灌服大肠埃希氏菌O78建立腹泻模型,分别用不同剂量郁金散改良组方、氟苯尼考及生理盐水治疗5 d(观察3 d),判定疗效.选取Ⅰ、Ⅱ、Ⅳ、Ⅴ、Ⅵ和Ⅷ试验组鸡只,测定血清生化指标、细胞因子含量,观察肝脏病理学和肠道变化.结果显示,治疗后药物组鸡只发热、精神沉郁、拉稀及糊肛等临床症状明显改善,观察期内无反复;与Ⅱ和Ⅷ组相比,Ⅴ组的平均日增重显著升高,料重比显著降低(P＜0.05);Ⅴ、Ⅵ、Ⅶ组治愈率、总有效率显著升高(P＜0.05),组间无差异(P＞0.05);血清ALT、AST、IL-6及TNF-α含量显著降低,ALB和IL-10含量显著升高(P＜0.05);Ⅴ和Ⅵ组心脏、肝脏、脾脏指数显著降低,法氏囊指数显著升高(P＜0.05),各药物组肝组织病理损伤均有明显改善.此外,Ⅴ组十二指肠绒隐比和大肠埃希氏菌载菌量极显著低于Ⅱ组(P＜0.01),显著低于Ⅷ组(P＜0.05).结果表明,郁金散改良组方可有效治疗鸡大肠埃希氏菌性腹泻,改善生长性能、组织损伤,抑制病原菌定植.综合疗效与成本分析,推荐2.7 g/kg为最佳给药剂量.
In order to determine the therapeutic effect and dosage of Yujin SAN Modified Formulaon chicken E.coli diarrhea.14 d Luhua chickens were divided into blank control group(Ⅰ),model control group(Ⅱ),1/3,1/2,1,2,3(Ⅲ、Ⅳ、Ⅴ、Ⅵ、Ⅶ)times TCV dose groups and flufenicol group(Ⅷ).In addition to Ⅰ,chickens in each group were treated with antibiotics and given Escherichia coli O78 to establish diarrhea model,and were treated with TCV,flufenicol and normal saline at different doses for 5 days(observation for 3 days)to determine the efficacy.Select Ⅳ,Ⅴ,Ⅵ,Ⅱ and Ⅷ test group chickens,serum biochemical in-dexes,cytokine content,liver pathology and intestinal changes were measured.The results demonstrated af-ter treatment,the clinical symptoms of fever,mental depression,diarrhea and anal paste of chickens in the drug group were significantly improved,and there was no recurrence during the observation period.Com-pared with Ⅱ and Ⅷ,the average daily gain of V was significantly increased,and the ratio of feed to gain was significantly decreased(P＜0.05).The cure rate and total effective rate of Ⅳ,Ⅴ and Ⅵ groups were significantly increased(P＜0.05),but there was no difference among groups(P＞0.05).The contents of ALT,AST,IL-6 and TNF-α in serum were significantly decreased,while the contents of ALB and IL-10 were significantly increased(P＜0.05).The indexes of heart,liver and spleen in Ⅴ and Ⅵ groups were sig-nificantly decreased,while bursa of Fabricius index was significantly increased(P＜0.05).The pathological injury of liver tissue was significantly improved in all drug groups.In addition,duodenal chow-to-crypto ra-tio and Escherichia coli load in Ⅴ group were significantly lower than those in Ⅱ group(P＜0.01),and significantly lower than those in Ⅷ group(P＜0.05).The result shows that TCV can effectively treat chicken Escherichia coli diarrhea,improve growth performance,tissue damage and inhibit pathogen coloni-zation.According to the comprehensive analysis of efficacy and cost,2.7 g/kg is recommended as the opti-mal dose.
