犊牛四种腹泻病毒多重RT-PCR检测方法的建立及应用OA
Establishment and Application of Multiplex RT-PCR Detection Methods for Four Diarrhea Viruses in Calves
为建立牛轮状病毒(BRV)、牛冠状病毒(BCoV)、牛病毒性腹泻病毒(BVDV)和牛纽布病毒(BNeV)的四重 PCR 检测方法,针对 BRV-VP6、BVDV-5'UTR、BCoV-NSP2、BNeV-DR BP 设计了特异性引物,经优化反应体系和条件,成功建立可同时检测BRV、BCoV、BVDV、BNeV的四重RT-PCR方法,并对云南部分地区采集的临床样本进行检测.结果显示该方法能特异性检出目标病毒,而BEV、BKV、BNoV等其他腹泻病原均呈阴性,具有良好的特异性.BR V、BCoV、BVDV、BNe V的最低检测限分别为1.34× 103、3.86× 103、1.06× 103、3.32× 103 copies/μL.对 322 份临床腹泻样品的检测结果,BRV 检出率 40.37%、BCoV检出率13.04%、BVDV检出率10.87%、BNeV检出率1.24%,BRV+BCoV混合感染检出率9.94%,BRV+BVDV检出率4.66%,BCoV+BVDV检出率为1.24%.四重RT-PCR方法和单一 RT-PCR方法符合率100%.该方法为犊牛病毒性腹泻病原的诊断、流行病学研究等提供了一种快捷简便的检测方法,临床样本检测结果BRV、BCoV、BVDV在腹泻犊牛中感染率较高,混合感染的情况突出.
To establish a quadruple PCR detection method for Bovine Rotavirus(BRV),Bovine Coronavirus(BCoV),Bovine Viral Diarrhea Virus(BVDV),and Bovine Nebovirus(BNeV),this study designed specific primers targeting BRV-VP6,BVDV-5'UTR,BCoV-NSP2,and BNeV-DR BP.By optimizing the reaction system and conditions,a quadruplex RT-PCR assay capable of simultaneously detecting BRV,BCoV,BVDV,and BNeV was successfully established and applied to clinical samples collected from various re-gions in Yunnan.Results demonstrated that this method specifically detected the target viruses,while other diarrheal pathogens such as BEV,BKV,and BNoV tested negative,indicating good specificity.The lower limits of detection(LOD)for BRV,BCoV,BVDV,and BNeV were 1.34 × 103 copies/μL,3.86 × 103 copies/μL,and 1.06 × 103 copies/μL,3.32 × 103 copies/μL,respectively.Testing of 322 clinical diarrheal samples revealed detection rates of 40.37%for BRV,13.04%for BCoV,10.87%for BVDV,and 1.24%for BNeV.Co-infection rates were 9.94%for BRV+BCoV,4.66%for BRV+BVDV,and 1.24%for BCoV+BVDV.The concordance rate between the quadruplex RT-PCR assay and single RT-PCR assays was 100%.This study provides a rapid and simple detection method for the diagnosis and epidemiological investigation of bovine viral pathogens.The detection results from clinical samples indicate high infection rates of BRV,BCoV,and BVDV in diarrheic calves,with the presence of co-infections.
