Multilocus Sequence Typing and Virulence Gene Analysis of Mycoplasma gallisepticum and Mycoplasma synoviae in Northern Fujian

为了解福建闽北地区鸡毒支原体(MG)和滑液囊支原体(MS)的种群结构、进化关系和毒力基因携带情况,根据PubMLST数据库公布的6个 MG管家基因(atpG、dnaA、fas A、rpoB、ruvB、uvrA)、7个MS管家基因(Adk、atpG、Efp、Gmk、nagC、Ppa、recA)作为分型参考,分别对闽北地区2023年2~10月间规模化养鸡场30份 MG核酸阳性病料和41份 MS核酸阳性病料进行测序和多位点序列分型(MLST),分析了不同菌株间的进化关系.结果显示,有6份MG阳性样本的6个管家基因全部扩增且测序成功,可分为3种ST型,分别为78、130、133,主要流行的ST型为ST-78(占50%),按照进化关系可分为A~D共4类,其主要流行株与疫苗株亲缘关系较近;有10份MS阳性样本的7个管家基因全部扩增且测序成功,可分为6种ST型,分别为43、80、93、96、147、151,主要流行的ST型为ST-147(占30%),按照进化关系可分为A~E 5类,其中只有2份ST-43型与MS-H株同在A类.毒力基因预测结果显示,闽北地区MG携带tuf、pdhB、vlh、gapA等10种毒力基因且以plpA为主要介导;MS携带tuf、plr/gap A、pdhB等7种毒力基因且以tuf和tnp为主要介导.研究结果丰富了福建闽北地区规模化鸡场鸡支原体流行病学数据,对鸡支原体感染的防控与净化提供参考.

To investigate the population structure,evolutionary relationships,and carriage of virulence genes of My-coplasma gallisepticum(MG)and Mycoplasma synoviae(MS)in the Fujian Minbei region,we used six MG housekeeping genes(atpG,dnaA,fasA,rpoB,ruvB,uvrA)and seven MS housekeeping genes(Adk,atpG,Efp,Gmk,nagC,Ppa,recA)from the PubMLST database as typing references.We performed sequencing and multilo-cus sequence typing(MLST)on 30 MG nucleic acid-positive samples and 41 MS nucleic acid-positive samples col-lected from large-scale chicken farms in Minbei between February and October 2023.The evolutionary relationships among different strains were analyzed.The results showed that among the 6 MG-positive samples,all 6 housekeep-ing genes were successfully amplified and sequenced,and they were classified into 3 sequence types(STs):ST78,ST130,and ST133.The predominant ST was ST78(50%),and according to evolutionary relationships,they were categorized into A-D4 groups,with the predominant strain being closely related to the vaccine strain.Among the 10 MS-positive samples,all 7 housekeeping genes were successfully amplified and sequenced,and they were classified into 6 STs:ST43,ST80,ST93,ST96,ST147,and ST151.The predominant ST was ST147(30%),and according to evolutionary relationships,they were categorized into A-E5 groups,with only 2 ST43 strains co-occurring with MS-H strains in group A.The virulence gene prediction results indicated that MG in the Minbei region carried 10 viru-lence genes,including tuf,pdhB,vlh,and gap A,with plp A as the main mediator.MS carried 7 virulence genes,including tuf,plr,gapA,and pdhB,with tuf and tnp as the main mediators.This study enriches the epidemiolog-ical data of mycoplasma in large-scale chicken farms in the Fujian Minbei region and provides a reference for the prevention,control,and eradication of mycoplasma infections in chickens.
