木芙蓉种间杂交后代的真实性鉴定OA
Authenticity Identification of Interspecific Hybrid Progeny in Hibiscus mutabilis
以木芙蓉'单瓣红''醉云'作为母本,樟叶槿、芙蓉葵作为父本进行杂交,共得到72株后代,利用亲本的叶片进行转录组测序筛选出差异性的SSR位点,并设计相应的引物进行扩增筛选,对木芙蓉种间杂交后代的真实性进行了鉴定,以期为其他植物的种间杂交鉴定提供参考依据.结果表明:筛选得到8对引物在亲本之间存在差异,在DG组合中,仅SSR020可对后代进行鉴定,并判定出DG-26为真杂种;DM及ZM组合中,有4对引物可对后代进行鉴定,在DM组合的8个后代中,仅DM-16无法判定是否为真杂种,其他7个均鉴定为真杂种,ZM组合中,SSR020引物在24个后代中鉴定出了6个后代为真杂种.利用表型进行交叉验证发现,鉴定出来的杂种确实表现出了亲本的中间形态,表明该研究筛选出来的分子标记真实有效,为木芙蓉和芙蓉葵的种间杂交提供了一些可利用的检测标记,对于缩短育种进程具有重要的意义.
Taking Hibiscus mutabilis'Danbanhong'and'Zuiyun'varieties as female parents and crossed them with Bombycidendron grewiifolium and H.moscheutos as male parents.A total of 72 progeny were obtained.Transcriptome sequencing of parental leaves was conducted to identify divergent SSR loci,and corresponding primers were designed for amplification and screening,in order to provide reference for the identification of interspecific hybridization of orther plants.The results showed that eight pairs of primers exhibited polymorphisms between the parents.In the DG combination,only SSR020 could authenticate the progeny,confirming DG-26 as a true hybrid.In the DM and ZM combinations,four pairs of primers were effective for progeny identification.Among the eight progeny in the DM combination,only DM-16 could not be definitively identified as a true hybrid,while the remaining seven were confirmed as true hybrids.In the ZM combination,SSR020 identified six out of 24 progeny as true hybrids.Phenotypic cross-validation demonstrated that the identified hybrids exhibited intermediate morphological traits between the parents,confirming the reliability and effectiveness of the molecular markers developed in this study.These markers provided valuable tools for interspecific hybridization between H.mutabilis and H.moscheutos,significantly shortening the breeding process.
