基于拮抗和多基因序列分析平菇种质资源遗传多样性OA
Genetic diversity of Pleurotus ostreatus germplasm resources based on antagonistic and multigene sequence analysis
[目的]本研究旨在结合拮抗和多基因序列分析技术,揭示供试平菇菌株种质资源的遗传变异程度和亲缘关系.[方法]以 15种平菇(Pleurotus ostreatus)菌株作为试验材料,首先通过拮抗试验测定各菌株间的拮抗反应,在 PDA平板上观察供试菌株的菌丝生长特性;同时提取供试菌株的基因组 DNA,扩增多基因序列构建系统进化发育树.[结果]金秀-2和金秀-1的菌丝生长速度为 4.722 mm·d⁻1,与其他菌株的差异达到极显著水平;香秀-1最慢,为 2.361 mm·d⁻1.大部分菌株之间存在不同程度的拮抗作用,只有金秀-2和金秀-1之间为无拮抗反应.ITS序列中菌株的总 GC 含量为 43.44%~46.71%,遗传距离变化范围为 0.002~0.397;TEFI-α 序列的总 GC 含量为52.06%~55.06%,遗传距离为 0.021~0.445;基于 ITS+TEFI-α 系统发育树、拮抗反应和生长速度结果,可将供试菌株 C、D、F、G、N、P 归为一类,菌株 A、E、H、I、J、L、M为一类,菌株 B分别单独为一类.[结论]本试验结合拮抗试验和多基因序列分析,明确了平菇种质资源存在较丰富的遗传多样性,菌株间遗传差异显著,为平菇种质资源的分类、鉴定及利用提供了依据.
[Objective]This study aimed to reveal the genetic variation and phylogenetic relationships of the tested Pleurotus ostreatus strains by combining cross-trial analysis with multi-gene sequence analysis techniques.[Method]Using 15 strains of P.ostreatus as test samples,the antagonistic interactions among strains were first determined through antagonism assays,and then the mycelial growth characteristics of the test strains on PDA plates were observed.Concurrently,genomic DNA was extracted from the test strains,and multiple gene se-quences were amplified to construct a phylogenetic tree.[Result]Mycelial growth characteristics revealed that the growth rates of Jinxiu-2 and Jinxiu-1 exhibited extremely significant differences compared to other strains,reach-ing to 4.722 mm·d⁻1.Xiangxiu-1 showed the slowest growth rate at 2.361 mm·d⁻1.Antagonistic interactions ex-isted to varying degrees among most strains,with only Jinxiu-2 and Jinxiu-1 exhibiting no antagonistic response.The total GC content in the ITS sequences ranged from 43.44%to 46.71%,with genetic distances varying from 0.002 to 0.397.The total GC content in the TEFI-α sequences ranged from 52.06%to 55.06%,with genetic dis-tances ranging from 0.021 to 0.445.Based on the ITS+TEFI-α phylogenetic tree,antagonistic response,and growth rate results,test strains C,D,F,G,N,and P formed one group,strains A,E,H,I,J,L,and M formed an-other group,and strain B stood alone as a separate group.[Conclusion]This study combined antagonism assays with polygenic sequence analysis to demonstrate that P.ostreatus germplasm resources exhibit substantial genetic diversity,with significant genetic differences among strains.These findings provide concrete evidence for the classification,identification,and utilization of P.ostreatus germplasm resources.
