目的:研究花叶饮代用茶对高脂血症模型大鼠的降脂作用及肠道菌群影响.方法:SD大鼠分为空白组、模型组及花叶饮低、中、高剂量组,高脂高糖饲料联合猪油灌胃建立高脂血症模型,造模成功后给药4周.检测血清总胆固醇(TC)、甘油三酯(TG)、低密度脂蛋白胆固醇(LDL-C)、高密度脂蛋白胆固醇(HDL-C)水平及天门冬氨酸氨基转移酶(AST)、丙氨酸氨基转移酶(ALT)活性;肝组织进行HE和油红O染色;16S rRNA测序分析肠道菌群改变.结果:与空白组比较,模型组体质量增长率、TC、TG、LDL-C水平及AST活性显著升高,HDL-C活性显著降低,肝组织出现明显脂肪变性与炎症,厚壁菌门、乳杆菌属、普雷沃氏菌属丰度降低,变形菌门、厌氧螺菌属丰度升高(均P<0.05或P<0.01).与模型组比较,花叶饮高剂量组第1周体质量增长率显著降低;各剂量组 TC、TG、LDL-C水平及AST活性显著降低,中剂量组 ALT显著降低,肝脂肪变性与炎症减轻;各剂量组厚壁菌门、经黏液真杆菌属丰度显著升高,高剂量组变形菌门、厌氧螺菌属丰度显著降低,中剂量组普雷沃氏菌属丰度显著升高(均P<0.05或P<0.01).相关性分析结果显示,乳杆菌丰度与HDL-C水平呈显著正相关(P<0.05);乳杆菌属、普雷沃氏菌属丰度与TC、TG、LDL-C水平、AST、ALT活性呈负相关趋势,与HDL-C水平呈正相关趋势;厌氧螺菌属则呈相反趋势.结论:花叶饮代用茶能降低高脂血症大鼠血脂,改善肝脏病理和肠道菌群失调,可用于高脂血症的日常预防和辅助治疗.

Objective:To investigate the lipid-lowering effect and intestinal flora modulation of Huayeyin herbal substitute tea in hyperlipidemic rats.Methods:SD rats were divided into blank group,model group,and low-,medium-and high-dose Huayeyin groups.The hyperlipidemia rat model was established by intragastric administration of high-fat and high-sugar diet com-bined with lard.After successful modeling,the rats were administered with corresponding interventions for 4 weeks.The serum levels of total cholesterol(TC),triglyceride(TG),low-density lipoprotein cholesterol(LDL-C)and high-density lipoprotein cho-lesterol(HDL-C),as well as the activities of aspartate aminotransferase(AST)and alanine aminotransferase(ALT)were detected.Liver tissues were stained with hematoxylin-eosin(HE)and Oil Red O.16S rRNA sequencing was performed to analyze the changes of intestinal flora.Results:Compared with the blank group,the weight growth rate,serum TC,TG,LDL-C levels and AST activity in the model group were significantly increased,while HDL-C level was significantly decreased.Obvious hepatic ste-atosis and inflammatory lesions were observed in liver tissues.The abundances of Firmicutes,Lactobacillus and Prevotella were decreased,whereas the abundances of Proteobacteria and Anaerospirillum were increased(all P<0.05 or P<0.01).Compared with the model group,the weight growth rate in the high-dose Huayeyin group was significantly reduced in the first week;the levels of TC,TG and LDL-C and the activity of AST were significantly decreased in each dose group,and ALT activity was significantly reduced in the medium-dose group,with alle-viated hepatic steatosis and inflammation.The abundances of Firmicutes and Blautia were significantly upregulated in all dose groups;the abundances of Proteobacteria and Anaerospi-rillum were markedly downregulated in the high-dose group,and the abundance of Prevotella was significantly elevated in the medium-dose group(all P<0.05 or P<0.01).Correlation analysis showed that the abundance of Lactobacillus was sig-nificantly positively correlated with HDL-C level(P<0.05).The abundances of Lactobacillus and Prevotella showed a negative correlation trend with TC,TG and LDL-C levels and AST and ALT activities,and a positive correlation trend with HDL-C level;while Anaerospirillum presented the opposite trend.Conclusion:Huayeyin herbal substitute tea lowers serum lipids,ameliorates liver pathology,and regulates gut dysbiosis in hyper-lipidemic rats.The results support its use in daily prevention and adjunctive treatment of hyperlipidemia.
