三棱-莪术有效组分配伍改善黄褐斑的作用研究OA
Study on the effect of active components compatibility of Sparganii Rhizoma and Curcumae Rhizoma in improving chloasma
目的:研究三棱与莪术有效成分配伍对黄褐斑改善作用的影响.方法:在体外细胞水平上,评估三棱总黄酮与莪术挥发油(1∶1)对α-MSH诱导的B16细胞内黑色素生成及酪氨酸酶活性的影响.随后,在体内动物水平上,通过黄体酮注射联合中波紫外线照射的方式构建C57BL/6小鼠耳廓黄褐斑模型,取血清检测SOD活性与MDA含量,并分别通过H&E染色和Masson Fontana染色观察耳廓组织病理学变化及黑色素沉积情况.结果:体外实验发现,与对照组比较,模型组细胞内酪氨酸酶活性及黑色素含量均显著上升(P<0.01).三棱总黄酮与莪术挥发油按质量比1∶1配伍能有效抑制该上升趋势(P<0.01),其效果最佳且呈明显浓度依赖性.在体内实验中,与正常组比较,模型组小鼠血清SOD活性下调(P<0.01),MDA水平上调(P<0.05),耳廓组织的黑色素分布增多(P<0.001);与模型组比较,三棱总黄酮与莪术挥发油配伍可改善各组小鼠耳廓组织中的黑色素分布(P<0.001),高剂量组上调 SOD活性(P<0.05)和下调MDA水平(P<0.05).结论:三棱总黄酮与莪术挥发油配伍具有改善黄褐斑作用,其机制可能与抑制黑色素生成及增强机体抗氧化能力有关.
Objective:To investigate the effect of the active component compatibility of Sparganii Rhizoma and Curcumae Rhizoma on improving chloasma.Methods:In vitro,the effects of total flavonoids from Sparganii Rhizoma and Curcumae Rhi-zoma oil on melanin production and tyrosinase activity in α-MSH-induced B16 cells were evaluated to determine the optimal com-patibility ratio.In vivo,a chloasma model was established on the auricles of C57BL/6 mice by progesterone injection combined with ultraviolet B irradiation.Serum samples were collected to detect superoxide dismutase(SOD)activity and malondialdehyde(MDA)content.Histopathological changes and melanin distribution in the auricular tissue were observed using hematoxylin-eosin(H&E)staining and Masson Fontana staining,respectively.Results:In vitro experiments showed that,compared with the control group,tyrosinase activity and melanin content were significantly increased in the model group(P<0.01).The compatibility of total flavonoids from Sparganii Rhizoma and Curcumae Rhizoma oil(1:1)effectively inhibited this increasing trend(P<0.01),with the most significant effect observed at this ratio and in a concentration-dependent manner.In vivo,compared with the normal group,the model group exhibited significantly decreased serum SOD activity(P<0.01),increased MDA level(P<0.05),and enhanced melanin distribution in the auricular tissue(P<0.001).Compared with the model group,treatment with the 1:1 compatibility of to-tal flavonoids from Sparganii Rhizoma and Curcumae Rhizoma oil improved melanin distribution in the auricular tissues of mice in all treatment groups(P<0.001),with the high-dose group significantly upregulating SOD activity(P<0.05)and downregulating MDA level(P<0.05).Conclusion:The compatibility of total flavonoids from Sparganii Rhizoma and Curcumae Rhizoma oil(1:1)has shown an ameliorative effect on chloasma,and its mechanism may be related to inhibiting melanin production and enhanc-ing antioxidant capacity.
