Determination of paeoniflorin in different batches of Moutan Cortex from different producing areas by HPLC
目的:测定不同产地不同批次牡丹皮中芍药苷含量.方法:首先建立并验证牡丹皮中芍药苷的HPLC测定方法,再选用市场上道地产区、量大产区的不同批次牡丹皮共计32批,本实验选用Waters Symmetry Shield C18色谱柱,其规格为250 mm×4.6 mm,粒径5 μm;流动相比例为16∶84,成分为乙腈与0.1%磷酸溶液;230 nm波长在检测时设定.样品前处理采用水回流提取法,提取时长为30 min,测定其中芍药苷含量,分析不同产地其中差异.结果:不同产地牡丹皮中芍药苷平均含量由高至低依次为安徽铜陵0.76%、安徽亳州0.73%、安徽南陵0.67%、山东菏泽0.75%、河南洛阳0.59%、重庆垫江0.66%、山西运城0.63%.结论:道地产区铜陵、主产区亳州、菏泽的牡丹皮中芍药苷平均含量较高.
Objective:To determinate paeoniflorin content in different batches of Moutan Cortex from different origins.Methods:Firstly,an HPLC method for the determination of paeoniflorin in Moutan Cortex was established and validated.A total of 32 batches of Moutan Cortex from authentic producing regions and large-yield producing regions on the market were selected.A Waters Symmetry Shield C18 column(250 mm×4.6 mm,5 μm)was used in this experiment.The mobile phase consisted of ace-tonitrile and 0.1%phosphoric acid solution at a ratio of 16∶84,and the detection wavelength was set at 230 nm.Samples were pre-treated by water reflux extraction for 30 min.The paeoniflorin content was determined,and the differences among samples from different producing regions were analyzed.Results:The average content of paeoniflorin in Moutan Cortex was 0.76%in Tongling,Anhui,0.73%in Bozhou,Anhui,0.67%in Nanling,Anhui,0.75%in Heze,Shandong,0.59%in Luoyang,Henan,0.66%in Dianjiang,Chongqing and 0.63%in Yuncheng,Shanxi.Conclusion:The average paeoniflorin contents of Moutan Cor-tex in Tongling,Bozhou and Heze main production areas are relatively higher.
