目的:基于核糖体内转录间隔区 2(ITS2)序列及二级结构,对市售前胡药材进行物种分析.方法:收集来自安徽、河北、广西等 5 个药材市场、11 个产区的 103 份市售前胡样品,通过提取DNA、聚合酶链式反应(PCR)扩增、双向测序获得ITS2 序列,利用MEGA 6.0 软件进行序列比对,计算鸟嘌呤和胞嘧啶占比与遗传距离,采用邻接法(NJ)构建系统聚类树,结合ITS2 数据库预测二级结构,进行物种鉴定.结果:前胡正品与其混伪品在NJ树上聚为独立分支;ITS2 二级结构在螺旋区上茎环大小、数目存在差异;市售前胡有 7 个基原,分别为白花前胡、石防风、短片拟藁本、华北前胡、当归、珊瑚菜和华中前胡.结论:基于ITS2 序列的DNA条形码及其二级结构能准确区分市售前胡药材正品及其混伪品.

Objective:To identify the species of commercial Peucedani Radix products based on the internal transcribed spacer 2(ITS2)sequence and its secondary structure.Methods:A total of 103 samples labelled as Peucedani Radix were collected from 11 producing regions distributed across five herbal markets in Anhui,Hebei,Guangxi and other provinces.Genomic DNA was extracted,and the ITS2 region was amplified by polymerase chain reaction(PCR)and bidirectionally sequenced.MEGA 6.0 was used for sequence alignment,and the guanine+cytosine(GC)content and genetic distances were calculated.A neighbour-joining(NJ)phylogenetic tree was constructed.The ITS2 secondary structure was predicted using the ITS2 database.Species identification was then performed.Results:Authentic Peucedani Radix and its adulterants formed separate clades in the NJ tree.The ITS2 secondary structures differed in both the size and number of stem-loops within the helical regions.Based on the NJ tree and secondary structure analysis,the commercial samples originated from seven distinct species:Peucedanum praeruptorum,P.terebinthaceum,Ligusticum brachylobum Franch.,P.harry-smithii var.subglabrum,Angelica sinensis,Glehnia littoralis,and P.medicum.Conclusion:The DNA barcode based on ITS2 combined with its secondary structure signature can reliably distinguish authentic Peucedani Radix from its adulterants.
