湖北恩施马铃薯病毒病流行与分布特征OA
Epidemiological characteristics and distribution patterns of potato viral diseases in Enshi Hubei
为明确湖北恩施马铃薯主产区病毒种类和发生情况.通过双抗体夹心酶联免疫吸附测定(DAS-ELISA)、二代测序(NGS)以及反转录聚合酶链式反应(RT-PCR)等分子技术对430份农户自留种薯进行了病毒检测.结果显示,马铃薯S病毒(potato virus S,PVS)和马铃薯M病毒(potato virus M,PVM)是构成该地区马铃薯病毒病害的主要病原,检出率分别为53.5%和23.0%,其他病毒种的检出率均低于10.0%.各病毒检出率随海拔改变而发生变化,马铃薯Y病毒(potato virus Y,PVY)在低海拔区域(<700 m,低山区)的检出率最高,达8.2%;马铃薯X病毒(potato virus X,PVX)在中海拔区域(700~1 000 m,二高山区)的检出率最高,为12.3%;PVS和PVM在高海拔区域(>1 000 m,高山区)的检出率分别高达64.2%和33.0%;马铃薯卷叶(potato leafroll virus,PLRV)和马铃薯A病毒(potato virus A,PVA)的检出率随海拔分布没有明显差异.马铃薯病毒流行图谱因地区而异,巴东和建始受PVS和PVM侵染最为严重,PVS检出率分别为90.6%和69.8%,PVM检出率分别为58.8%和34.0%.PVX和PVA在鹤峰更流行,检出率分别为20.0%和35.0%.恩施马铃薯病毒侵染模式以单一和双重侵染为主,分别占侵染样本数的52.8%和37.8%,而三重和四重侵染的发生率较低,仅占8.7%和0.7%.此外,恩施部分地区存在马铃薯H病毒(potato virus H,PVH)和马铃薯帚顶病毒(potato mop-top virus,PMTV)的轻微侵染.本研究为恩施地区马铃薯病毒病的防控提供了重要的基础信息.
To clarify the types and occurrence of viruses in the main potato production areas of Enshi,virus detection was conducted on 430 self-retained potatoes from farmers using double antibody sandwich-enzyme linked immunosorbent assay(DAS-ELISA),next-generation sequencing(NGS)and reverse transcription-polymerase chain reaction(RT-PCR).The results showed that potato virus S(PVS)and potato virus M(PVM)were the main pathogens causing potato virus diseases in this region.The detection rates of PVS and PVM were 53.5%and 23.0%respectively,and the detection rates of other virus species were all below 10.0%.The detection rate of viruses varies with changes in altitude.Potato virus Y(PVY)had the highest detection rate of 8.2%in low-altitude areas(<700 m),potato virus X(PVX)had the highest detection rate of 12.3%in mid-altitude areas(700-1 000 m),PVS and PVM had detection rates as high as 64.2%and 33.0%,respectively,in high-altitude areas(>1 000 m).Potato leafroll virus(PLRV)and potato virus A(PVA)showed no significant difference in distribution across altitudes.The epidemiological patterns of potato viruses vary from region to region.Badong and Jianshi were most severely infected by PVS and PVM,with PVS detection rates of 90.6%and 69.8%,respectively,and PVM detection rates of 58.8%and 34.0%,respectively.PVX and PVA were more prevalent in Hefeng,with detection rates of 20.0%and 35.0%,respectively.The main patterns of potato virus infection in Enshi were single and double infections,accounting for 52.8%and 37.8%of the infected samples,respectively,while triple and quadruple infections occurred less frequently,accounting for only 8.7%and 0.7%,respectively.Additionally,mild infections of potato virus H(PVH)and potato mop-top virus(PMTV)were present in some areas of Enshi.This study provides important basic information for prevention and control of potato viruses in the Enshi region.
