云南丽江金沙江流域优质烟区番茄斑萎病毒中间寄主调查与鉴定OA
Investigation and identification of intermediate hosts of tomato spotted wilt virus in high-quality tobacco-growing areas of the Jinsha River Basin in Lijiang,Yunnan province
番茄斑萎病毒(tomato spotted wilt virus,TSWV)是正番茄斑萎病毒属Orthotospovirus成员,为负义链RNA病毒,常常对番茄、辣椒以及烟草等经济作物造成严重危害.2021年—2023年,在云南丽江金沙江流域烟草种植区内采集疑似感染TSWV的烟草样品33份和杂草样品219份,利用免疫金标速测卡对采集的样品进行TSWV检测,其中24份烟草样品为阳性,检出率为72.72%;89份杂草样品为阳性,检出率为40.63%,阳性杂草寄主分属13科,21属,共24种,其中半夏和蜜蜂花为TSWV新寄主.通过电子显微镜观察,从感病的烟草和鬼针草上均发现球形病毒粒体,大小80~90 nm,具有正番茄斑萎病毒属病毒的典型形态特征.通过RT-PCR检测、克隆、测序,共获得19条TSWV N基因序列;序列相似性分析发现,该19条TSWV N基因序列的核苷酸相似性及与其他TSWV分离物的N基因序列相似性均在97.8%~99.9%.系统进化分析发现,本研究获得的19条TSWV N基因序列与TSWV中国分离物聚于一个分支,亲缘关系较近.种群遗传多样性分析发现,TSWV杂草种群的遗传多样性高于TSWV烟草种群.本研究结果有助于深入了解云南丽江金沙江流域TSWV寄主种类和发生流行规律,为综合防控该地区TSWV侵染引起的病害提供理论依据.
Tomato spotted wilt virus(TSWV)belongs to the genus Orthotospovirus and is a negative-sense RNA virus that often causes serious damage to economically important crops such as tomato,pepper and tobacco.During 2021-2023,33 tobacco and 219 weeds leaf samples suspected of TSWV infection were collected from tobacco-growing areas in the Jinsha River Basin of Lijiang,Yunnan province.Using a colloidal gold immunochromatographic rapid test kit,TSWV infection was detected in 24 tobacco samples(72.72%)and 89 weed samples(40.63%).The positive weed hosts belonged to 13 families,21 genera and 24 species,among which Pinellia ternata and Melissa axillaris were identified as new hosts of TSWV.Electron microscopy observation revealed spherical virions with diameters of approximately 80-90 nm in infected tobacco and Bidens pilosa,exhibiting typical morphological characteristics of viruses in the genus Orthotospovirus.Through RT-PCR detection,cloning,and sequencing,a total of 19 TSWV N gene sequences were obtained.Sequence similarity analysis showed that the nucleotide identities of these 19 N gene sequences and their identities with TSWV N gene sequences from other isolates ranged from 97.8%to 99.9%.Phylogenetic trees showed that the 19 TSWV N gene sequences obtained in this study clustered with TSWV isolates from China and showed close genetic relationships.Population genetic diversity analysis revealed that the genetic diversity of the TSWV weed population was higher than that of the TSWV tobacco population.These results contribute to a better understanding of host species and the occurrence and epidemic patterns of TSWV in the Jinsha River Basin of Lijiang,Yunnan province,and provide a theoretical basis for the integrated management of diseases caused by TSWV infection in this region.
