基于UFLC-Q-TOF-MS/MS技术分析障眼明片浸膏的化学成分和入血成分OA
Analysis of chemical components and blood plasma constituents of Zhangyanming Tablet extract based on UFLC-Q-TOF-MS/MS method
基于超快速液相色谱-四极杆飞行时间质谱(UFLC-Q-TOF-MS/MS)技术全面研究障眼明片浸膏的化学成分和入血成分.以水(含φ=0.1%甲酸)-乙腈(含φ=0.1%甲酸)为流动相进行梯度洗脱,化学成分和入血成分分别采用 Hypersil GOLD 色谱柱(4.6 mm×250 mm,5 μm)和 Kinetex C18色谱柱(2.1 mm×100 mm,2.6 μm)进行分离,流速分别为1.0和0.3 mL/min,柱温分别为35和40 ℃;采用电喷雾离子源(ESI)正、负模式扫描,范围为m/z 50~1 500.通过分析保留时间、加合离子峰、二级质谱碎片,结合对照品、数据库和文献比对,共在障眼明片浸膏中鉴定出200个化学成分,包括47种黄酮类、41种萜类、31种有机酸类、20种苯丙素类、17种生物碱类、10种糖类、8种氨基酸类、4种蒽醌类、4种苯酞类、3种香豆素类及15种其他类;在Beagle犬含药血浆中共鉴定出46种化学成分,包括39种原型化合物和7种代谢成分.本研究对障眼明片浸膏的化学成分和入血成分进行了系统、快速的分离鉴定,并总结其主要化合物类型的质谱裂解规律,为其药效物质基础和质量评价研究提供了科学依据与参考.
To comprehensively investigate the chemical components and constituents in plasma of Zhangyanming Tablet extract using ultra-fast liquid chromatography-quadrupole-time-of-flight tandem mass spectrometry(UFLC-Q-TOF-MS/MS).Gradient elution was performed using mobile phase consisting of water and acetonitrile,both containing φ=0.1%formic acid.Separation of chemical components and constituents in plasma was conducted on Hypersil GOLD(4.6 mm×250 mm,5 μm)and Kinetex C18(2.1 mm×100 mm,2.6 μm)column,with flow rate of 1.0 and 0.3 mL/min,and column temperature maintained at 35 and 40 ℃,respectively.Electrospray ionization(ESI)source was used in both positive and negative ion mode,with a scanning m/z range from 50 to 1 500.Through analyzing retention time,adduct ion peak,MS/MS fragments,and comparison with reference standards,database,and literature data,a total of 200 chemical compounds were identified in Zhangyanming Tablet extract,including 47 flavonoids,41 terpenoids,31 organic acids,20 phenylpropanoids,17 alkaloids,10 saccharides,8 amino acids,4 anthraquinones,4 phthalides,3 coumarins and 15 other components.In Beagle dog plasma,a total of 46 chemical constituents were detected,among which 39 were prototype compounds and 7 were metabolites.This study established a systematic and efficient approach for the separation and identification of chemical components and constituents in plasma of Zhangyanming Tablet extract.The study summarized the characteristic fragmentation patterns of major compound types,providing scientific evidence and references for investigating the pharmacologically active substances and quality evaluation of Zhangyanming Tablet extract.
