山茱萸提取物缓解小鼠急性肾损伤的功能活性评价OA
Evaluation of the functional activity of Cornus officinalis extract in alleviating acute kidney injury in mice
为探究山茱萸提取物缓解急性肾损伤的潜在机制,本研究选取 C57BL/6J 雄性小鼠,通过腹腔注射庆大霉素建立小鼠急性肾损伤模型,将小鼠分为正常对照组、模型组及山茱萸提取物干预组,分别检测小鼠体重、脏器系数、血清肌酐与尿素氮浓度、炎症因子质量浓度,观察肾脏组织病理变化,并分析肠道菌群组成.结果显示,与模型组相比,山茱萸提取物组小鼠体重变化率有所下降;血清肌酐和尿素氮浓度显著降低;IL-6与 TNF-α 质量浓度显著下降,IL-10 质量浓度显著升高;肾小管上皮细胞坏死、炎症浸润及纤维化等病理损伤明显缓解;肠道中有益菌丰度增加、有害菌丰度减少.结果表明,山茱萸提取物可通过调节炎症反应及改善肠道微生态,对急性肾损伤发挥保护作用.本研究从组织学、免疫学和微生态学角度阐明了山茱萸提取物对急性肾损伤的影响,为山茱萸提取物的功能评价与精准应用提供了参考.
To investigate the potential mechanism by which Cornus officinalis extract alleviates acute kidney injury,male C57BL/6J mice were selected.An acute kidney injury model was established in mice via intraperitoneal injec-tion of gentamicin.The mice were divided into a normal control group,a model group,and a Cornus officinalis ex-tract intervention group.Body weight,organ coefficients,serum creatinine and urea nitrogen concentrations,and in-flammatory cytokine mass concentrations were measured.Renal histopathological changes were observed by tissue sectioning,and the composition of the intestinal flora was analyzed.The results showed that,compared with the mod-el group,the rate of body weight change was lower in the Cornus officinalis extract group;serum creatinine and urea nitrogen concentrations were significantly reduced;IL-6 and TNF-α mass concentrations were significantly decreased,while IL-10 mass concentration was significantly increased;pathological injuries such as renal tubular epithelial cell necrosis,inflammatory infiltration,and fibrosis were significantly alleviated;the abundance of beneficial bacteria in the intestine increased,while the abundance of harmful bacteria decreased.The results indicate that Cornus officina-lis extract can exert a protective effect on acute kidney injury by regulating inflammatory responses and improving the intestinal microecology.This study elucidates the effects of Cornus officinalis extract on acute kidney injury from his-tological,immunological,and microbiological perspectives,providing a reference for the functional evaluation and precise application of Cornus officinalis extract.
