[目的]研究尼罗罗非鱼(Oreochromis niloticus)免疫后特异性IgM抗体水平与抗无乳链球菌(Streptococ-cus agalactiae)感染能力的关联性,估算该抗病性状的遗传参数,以评估其作为抗病育种指标的潜力.[方法]采用无乳链球菌灭活疫苗浸泡免疫3个尼罗罗非鱼群体(花都、番禺和海南群体),于初次免疫后15 d按相同免疫程序加强免疫1次,分别于免疫前(0 d)、免疫后7、14、21、28、60、90 d检测免疫后血清特异性IgM抗体水平,揭示其动态变化规律,并分析免疫后血清IgM抗体峰值水平与罗非鱼人工感染无乳链球菌后的免疫保护率、存活情况及血液载菌量的相关性.通过构建多个全同胞家系,检测各家系子代免疫后特异性IgM抗体水平,采用约束极大似然法(REML)估算其方差组分和遗传力,并运用最佳线性无偏预测(BLUP)法预测个体育种值.[结果]罗非鱼免疫后血清特异性IgM抗体水平于第2次免疫后7 d达到峰值(68.07~91.73 μg/mL),并维持其高质量浓度达28 d(>54.99 μg/mL).免疫后IgM抗体水平越高的群体,其相对免疫保护率也越高;攻毒后存活个体的IgM抗体免疫应答水平均显著高于死亡个体(P<0.05),且IgM抗体免疫应答水平越高的个体攻毒后存活时间越长.对未免疫的罗非鱼进行攻毒,其血清中特异性IgM水平与血液中无乳链球菌含量呈极显著负相关(P<0.01).基于24个全同胞家系的遗传分析表明,罗非鱼IgM抗体免疫应答性状的遗传力为0.44.[结论]罗非鱼IgM抗体免疫应答水平与其抗无乳链球菌感染能力呈正相关,且属于高遗传力性状.IgM抗体免疫应答水平可作为罗非鱼抗病选育的重要候选性状,为培育易免疫抗病新品系奠定了理论基础.

[Objective]This study aims to elucidate the correlation between specific IgM antibody levels and resistance to Streptococcus agalactiae infection in tilapia(Oreochromis niloticus)after immunization,and estimate the genetic parameters of this disease resistance trait,and evaluate its poten-tial as an indicator for disease-resistant breeding.[Method]Three populations of tilapia(Huadu,Panyu,and Hainan populations)were immunized by immersion with an inactivated S.agalactiae vac-cine,followed by a booster immunization at 15 days post-initial immunization(dpi).The levels of serum-specific IgM antibody were detected prior to immunization(0 d)and at 7,14,21,28,60,and 90 dpi to reveal its temporal dynamics.The correlations between the peak levels of serum-specific IgM antibody and relative percent survival(RPS),survival status and blood bacterial load following artifi-cial challenge with S.agalactiae were analyzed.Multiple full-sib families were constructed,and serum-specific IgM antibody levels of offspring from each family after immunization were measured.The vari-ance components and heritability were estimated using restricted maximum likelihood(REML),and the individual breeding values were predicted by best linear unbiased prediction(BLUP).[Result]The serum-specific IgM antibody levels in tilapia peaked(from 68.07 to 91.73 μg/mL)at 7 days after the second immunization and persisted at a high mass concentration(>54.99 μg/mL)for 28 days.Tilapia populations with higher post-immunization IgM antibody levels exhibited higher relative percent sur-vival(RPS).Surviving individuals after the bacterial challenge showed significantly higher IgM anti-body response levels than those that died(P<0.05),and individuals with higher IgM antibody response levels had a longer survival time after the challenge.In non-immunized tilapia subjected to bacterial challenge,serum-specific IgM levels were extremely significantly negatively correlated with blood S.agalactiae load(P<0.01).Genetic analysis based on 24 full-sib families revealed that the heritability of the IgM antibody immune response trait in tilapia was 0.44.[Conclusion]IgM antibody immune response level in tilapia is positively correlated with its resistance to S.agalactiae infection and repre-sents a highly heritable trait.The IgM antibody immune response level can serve as an important candi-date trait for disease-resistant breeding in tilapia,providing a theoretical foundation for developing new strains with enhanced immunocompetence and disease resistance.
