[目的]探究不同中草药饲料添加剂对珍珠龙胆石斑鱼(Epinephelus fuscoguttatus ♀×Epinephelus lanceolatus ♂)生长性能、血清生理生化指标、肠道消化酶活性、肝脏抗氧化免疫功能及肠道菌群结构的影响,为中草药饲料添加剂在珍珠龙胆石斑鱼健康养殖中的应用提供理论依据.[方法]以初始体质量为(17.64±0.20)g 的珍珠龙胆石斑鱼为研究对象,设置对照组、1%(质量分数,下同)冬凌草(Rabdosiae rubescentis herba)组、2%冬凌草组、1%冬凌草复方组、2%冬凌草复方组、1%逍遥散组和2%逍遥散组共7个处理组,每组3个重复,每个重复15尾鱼,进行56 d养殖实验.测定鱼体生长性能、形体指标、血清生理生化指标、肠道消化酶活性、肝脏抗氧化及非特异性免疫指标,并采用16S rRNA高通量测序分析肠道菌群组成.[结果]与对照组相比,1%冬凌草组、1%冬凌草复方组、2%冬凌草复方组和1%逍遥散组终末均质量、增质量率和特定生长率显著升高,各中草药添加组丙氨酸氨基转移酶和天冬氨酸氨基转移酶活性均显著低于对照组.1%冬凌草复方组、2%冬凌草复方组、1%逍遥散组、2%逍遥散组和2%冬凌草组组淀粉酶活性显著高于1%冬凌草组和对照组,2%冬凌草复方组胃蛋白酶活性显著高于1%冬凌草组、2%冬凌草组、1%逍遥散组和对照组.1%冬凌草组、2%冬凌草组和2%逍遥散组过氧化氢酶活性显著升高,2%冬凌草复方组和2%逍遥散组超氧化物歧化酶活性显著高于其他处理组;2%冬凌草复方组丙二醛含量显著低于对照组.中草药添加组肠道菌群丰富度总体高于对照组,且各组菌群结构存在差异;在属水平上,逍遥散组乳酸球菌(Lactococcus)属丰度较高,冬凌草组芽孢杆菌属(Bacillus)丰度较高,1%冬凌草组、1%冬凌草复方组、2%冬凌草复方组、1%逍遥散组和2%逍遥散组弧菌属(Vibrio)丰度显著低于对照组.α=0.05.[结论]在本实验条件下,添加质量分数为2%的冬凌草复方中草药综合效果最佳,可作为促进珍珠龙胆石斑鱼健康养殖的潜在绿色饲料添加剂.

[Objective]To explore the effects of different Chinese herbal medicine feed additives on growth performance,serum physiological and biochemical indexes,intestinal digestive enzyme activity,liver antioxidant immune function and intestinal flora structure of pearl gentian grouper(Epinephelus fuscoguttatus♀×Epinephelus lanceolatus♂),and to provide theoretical basis for the application of Chinese herbal medicine feed additives in the healthy breeding of pearl gentian grouper.[Method]A feeding trial lasting 56 days was conducted using the pearl gentian grouper with initial body mass of(17.64±0.20)g as the research object,and seven treatment groups were set up,including control group,1%(implying mass fraction in feed,the same below)Rabdosiae Rubescentis Herba(Rabdosia rubescens)group,2%R.rubescens group,1%R.rubescens compound group,2%R.rubescens compound group,1%Xiaoyao San group and 2%Xiaoyao San group,with three replicates in each group and 15 fish in each replicate.The growth performance,body shape index,serum physiological and biochemical indexes,intestinal digestive enzyme activity,liver antioxidant and non-specific immune indexes of the fish were measured,and the composition of intestinal flora was analyzed by 16S rRNA high-throughput sequencing.[Result]Different Chinese herbal medicine additives had significant effects on the growth performance,digestive ability,antioxidant immune status and intestinal flora structure of pearl gentian grouper.Compared with the control group,the final average mass,mass gain rate and specific growth rate of 1%R.rubescens group,1%R.rubescens compound group,2%R.rubescens compound group and 1%Xiaoyao San group were significantly increased.Serum biochemical indicators showed that the activities of alanine aminotransferase and aspartate aminotransferase in each Chinese herbal medicine addition group were significantly lower than those in the control group.The results of digestive enzyme showed that the amylase activity of 1%R.rubescens compound group,2%R.rubescens compound group,1%Xiaoyao San group,2%Xiaoyao San group and 2%R.rubescens group was significantly higher than that of 1%R.rubescens group and control group.The pepsin activity of 2%R.rubescens compound group was significantly higher than that of 1%R.rubescens group,2%R.rubescens group,1%Xiaoyao San group and control group.The liver antioxidant and immune indexes showed that the catalase activity of 1%R.rubescens group,2%R.rubescens group and 2%Xiaoyao San group were significantly increased.The activity of superoxide dismutase in 2%R.rubescens compound group and 2%Xiaoyao San group were significantly higher than that in other treatment groups.At the same time,the content of malondialdehyde in the 2%R.rubescens compound group was significantly lower than that in the control group.The analysis of intestinal flora showed that the richness of intestinal flora in the Chinese herbal medicine addition groups were higher than that in the control group,and there were differences in the structure of intestinal flora in each group.At the genus level,the abundance of Lactococcus in Xiaoyao San group was higher,the abundance of Bacillus in R.rubescens group was higher,and the abundance of Vibrio in 1%R.rubescens group,1%R.rubescens compound group,2%R.rubescens compound group,1%Xiaoyao San group and 2%Xiaoyao San group were significantly lower than that in control group.α=0.05.[Conclusion]Under the conditions of this experiment,the comprehensive effect of adding 2%R.rubescens compound Chinese herbal medicine is the best,which can be used as a potential green feed additive to promote the healthy breeding of pearl gentian grouper.
