活菌数含量是评价布鲁氏菌病活疫苗免疫效力的关键指标.通过建立科学的不确定度评定方法,不仅有助于疫苗生产企业优化生产工艺、控制产品质量,也为监管部门制定更合理的质量标准提供了技术支撑.本研究对活菌计数法测定布鲁氏菌病活疫苗活菌数含量结果的不确定度进行了评定.采用《中华人民共和国兽药典(2020 年版)》活菌(芽孢)计数法对布鲁氏菌病活疫苗产品的活菌数含量进行测定.依据《合格评定 生物样本测量不确定度评定与表示应用指南》的相关规范,对活菌计数法测定过程可能引入的各类不确定度因素进行分析、评定和量化.计算确认活菌计数法测定布鲁氏菌病活疫苗活菌数含量的扩展不确定度(U)为5.6302(包含因子 k=2),其不确定度主要来源为培养箱温度控制和吸管操作误差.

Live bacterial counting is a critical indicator for evaluating the immunological efficacy of brucellosis vaccines.Establishing a scientific uncertainty evaluation method not only helps vaccine manufacturers optimize production processes and control product quality,but also provides technical support for regulatory authorities to formulate more rational quality standards.In this study,the uncertainty associated with the determination of live bacterial count in brucellosis vaccines using the live bacterial counting was evaluated.The live bacterial count of brucellosis vaccine was determined in accordance with the Live Bacteria(Spore)Counting of the Veterinary Pharmacopoeia of the People's Republic of China(2020 Edition).Following the relevant provisions of the Conformity assessment—Application guide for evaluation and expression of uncertainty in biological sample measurement,various uncertainty factors potentially introduced during the determination process were analyzed,evaluated,and quantified.The calculated expanded uncertainty(U)for the determination was 5.6302(coverage factor k=2),and the main factors of uncertainty were identified as incubator temperature control and pipetting operation errors.
