目的 基于代谢组学探讨青盲一号方干预外伤性视神经病变(TON)模型大鼠的作用机制.方法 大鼠随机分为对照组(CG)、模型组(MG)和青盲一号方组(QM),每组6只.MG组、QM组采用视神经横向牵拉法建立TON模型,CG组仅暴露视神经,不进行牵拉.QM组每日灌胃13 g/kg青盲一号方,CG组、MG组灌胃等体积蒸馏水,连续给药14 d.给药完成后取大鼠视网膜,采用超高效液相色谱系统、高分辨液质联用仪检测样品,并进行代谢组学信息分析.结果 (1)代谢物数据库注释及功能分类:代谢物的分类主要为脂类及类脂分子、有机酸及衍生物、有机氧化合物等,相关代谢通路主要涵盖癌症、神经退行性疾病、消化系统、内分泌系统、神经系统和感觉系统等.(2)差异代谢物分析:CG组与MG组大鼠视网膜共检出差异代谢物1,121个,其中上调700个、下调421个;QM组与MG组大鼠视网膜共检出差异代谢物1,153个,其中上调528个、下调625个.进一步分析共鉴定出37种差异代谢物.(3)代谢通路分析:显著相关的代谢通路有甘油磷脂代谢、花生四烯酸代谢、糖基磷脂酰肌醇(GPI)锚定生物合成;而醚脂代谢、类固醇激素生物合成及不饱和脂肪酸的生物合成通路的显著性与影响力较低.结论 青盲一号方主要通过调控甘油磷脂代谢、花生四烯酸代谢及GPI锚定生物合成发挥抗炎、抗氧化、稳定细胞膜结构与保护神经功能等多重效应,从而减轻视神经损伤,促进视觉功能恢复.

OBJECTIVE To explore the mechanism of Qingmang Yihao Formula in intervening in traumatic optic neuropathy(TON)model rats based on metabolomics.METHODS Rats were randomly divided into a control group(CG),a model group(MG),and a Qingmang Yihao Formula group(QM),with six rats in each group.The MG and QM groups underwent establishment of the TON model using the lateral optic nerve traction method,while the CG group only had the optic nerve exposed without traction.The QM group was intragastrically administered 13 g/kg of Qingmang Yihao Formula daily,and the CG and MG groups were administered an equal volume of distilled water,for a continuous administration period of 14 days.After administration,rat retinas were collected.Samples were detected using an ultra-high performance liquid chromatography system and a high-resolution liquid chromatography-mass spectrometer,followed by metabolomic information analysis.RESULTS(1)Metabolite database annotation and functional classification:The classification of metabolites mainly included lipids and lipid-like molecules,organic acids and derivatives,and organic oxygen compounds.The related metabolic pathways primarily covered cancer,neurodegenerative diseases,the digestive system,endocrine system,nervous system,and sensory system.(2)Differential metabolite analysis:A total of 1,121 differential metabolites were detected in rat retinas between the CG and MG groups,with 700 upregulated and 421 downregulated.Between the QM and MG groups,1,153 differential metabolites were detected,with 528 upregulated and 625 downregulated.Further analysis identified 37 differential metabolites.(3)Metabolic pathway analysis:The significantly related metabolic pathways were glycerophospholipid metabolism,arachidonic acid metabolism,and glycosylphosphatidylinositol(GPI)-anchor biosynthesis.While the significance and impact of ether lipid metabolism,steroid hormone biosynthesis,and biosynthesis of unsaturated fatty acids pathways were lower.CONCLUSIONS Qingmang Yihao Formula primarily exerts multiple effects such as anti-inflammatory,antioxidant,stabilization of cell membrane structure,and protection of neural function by regulating glycerophospholipid metabolism,arachidonic acid metabolism,and GPI-anchor biosynthesis,thereby alleviating optic nerve injury and promoting the recovery of visual function.
