芷皂烧伤膏对大鼠深Ⅱ度烧烫伤的创面修复作用研究OA
Study on Wound Repair Effect of Zhizao Burn Ointment on Deep Second Degree Scalds in Rats
目的 观察芷皂烧伤膏对大鼠深Ⅱ度烧烫伤创面的修复作用.方法 取SD大鼠15只,制作深Ⅱ度烧烫伤动物模型,随机分为3组,每组5只,对照组予生理盐水湿敷创面,凡士林油纱覆盖;芷皂烧伤膏组及湿润烧伤膏组分别予药膏外敷创面,治疗20 d.于造模当天及实验开始后第3、7、10、14、20日观察创面愈合情况,计算创面愈合率.于第20日尾静脉取血,检测血常规及肝肾功能,随后处死动物,以烧伤中心为原点取皮肤组织,苏木精-伊红(HE)染色后观察组织形态,免疫组化法观察Ki67、B细胞淋巴瘤2(Bcl-2)表达.结果 第7日至第20日,芷皂烧伤膏组未愈合面积、创面愈合率均小于对照组(P<0.01),在第10日亦高于湿润烧伤膏组(P<0.01);第7日至第20日,芷皂烧伤膏组痂皮面积小于对照组(P<0.05),在第10日,芷皂烧伤膏组痂皮面积亦小于湿润烧伤膏组(P<0.05).第20日,芷皂烧伤膏组新生表皮组织结构更接近正常皮肤,其厚度低于对照组及湿润烧伤膏组(P<0.01),炎细胞浸润数低于对照组(P<0.01).芷皂烧伤膏组与湿润烧伤膏组表皮基底层Ki67细胞、Bcl-2细胞阳性率均明显低于对照组(P<0.05),芷皂烧伤膏组Bcl-2细胞阳性率低于湿润烧伤膏组(P<0.05).芷皂烧伤膏组及湿润烧伤膏组血常规及肝肾功指标与对照组差异无统计学意义(P>0.05).结论 芷皂烧伤膏能促进深Ⅱ度烧烫伤创面快速愈合,促进痂皮脱落,抑制愈后皮肤纤维组织增生,抑制瘢痕生成,无明显不良反应.
Objective:To investigate the wound repair effect of Zhizao Burn Ointment on deep second degree scalds in rats.Methods:Fifteen SD rats were selected to establish deep second degree scald animal models and randomly divided into 3 groups with 5 rats in each group.The control group was treated with normal saline wet compress on wounds and covered with vaseline gauze stripes,the Zhizao Burn Ointment group and Moist Burn Ointment group were treated with external ointment application respectively for 20 days.Wound healing was ob-served on the day of modeling and the 3rd,7th,10th,14th and 20th day after treatment,and the wound healing rate was calculated.Blood was collected from the tail vein at the 20th day to detect routine blood test,liver and kidney functions.Then the animals were sacrificed,and skin tissue was taken from the center of the burn as the or-igin.Histomorphology was observed after HE staining,and the expressions of Ki67 and Bcl-2 were observed by im-munohistochemistry.Results:From the 7th to 20th day,the unhealed area and wound healing rate in the Zhizao Burn Ointment group were lower than those in the control group(P<0.01),and were also higher than those in the Moist Burn Ointment group at the 10th day(P<0.01).From the 7th to 20th day,the eschar area in the Zhizao Burn Ointment group was smaller than that in the control group(P<0.05),and was also smaller than that in the Moist Burn Ointment group at the 10th day(P<0.05).At the 20th day,the histological structure of the newborn epidermis in the Zhizao Burn Ointment group was closer to normal skin,with thickness lower than that in the con-trol group and Moist Burn Ointment group(P<0.01),and inflammatory cell infiltration count lower than that in the control group(P<0.01).The positive rates of Ki67 cells and Bel-2 cells in the epidermal basal layer in the Zhizao Burn Ointment group and Moist Burn Ointment group were significantly lower than those in the control group(P<0.05),and the positive rate of Bcl-2 cells in the Zhizao Burn Ointment group was lower than that in the Moist Burn Ointment group(P<0.05).There were no significant differences in routine blood test,liver and kidney function indexes between the Zhizao Burn Ointment group,Moist Burn Ointment group and the control group(P>0.05).Conclusion:Zhizao Burn Ointment can promote rapid healing of deep second degree scald wounds,acceler-ate eschar shedding,inhibit skin fibrous tissue hyperplasia after healing and scar formation,with no obvious ad-verse reactions.
