基于UPLC-MS/MS的临床验方愈疮散化学成分分析OA
Chemical Composition Analysis of Clinical Empirical Formula Yuchuang Powder Based on UPLC-MS/MS
目的:建立超高效液相色谱-串联质谱法(UPLC-MS/MS)系统分析临床验方愈疮散的化学成分,解析代表性化合物的裂解规律,提升中药制剂的质量标准.方法:采用 UPLC-MS/MS 鉴定愈疮散中的化学成分.色谱条件如下,色谱柱为 ACQUITY UPLC HSS T3 柱(100 mm×2.1 mm,1.8 μm),流动相为 0.1%甲酸水溶液-乙腈(梯度洗脱),进样量为 10 μL.基于 UNIFI 中药数据库,应用 UNIFI 软件进行自动筛查,对照公共数据库及已发表文献对化学成分进行分类、鉴定,碎片离子匹配.结果:共鉴定出80 个化学成分,其中正离子模式检出 58 个,负离子模式检出 41 个,19 个成分在 2 种离子模式下均被检出.按化合物类别划分,主要包括萜类 39 个、菲类7 个、酚酸类8 个、生物碱类4 个,以及黄酮类、维生素 D 衍生物、脂肪酸类等成分.结论:本研究建立的UPLC-MS/MS 法可鉴定愈疮散中具有药理活性的化学成分和各个药材的特征性成分,为该制剂化学成分的快速鉴定及质量控制提供参考.
OBJECTIVE:To establish an ultra-performance liquid chromatography-tandem mass spectrometry(UPLC-MS/MS)system for the comprehensive analysis of the chemical components of clinical empirical formula Yuchuang powder,to elucidate the fragmentation patterns of representative compounds,and to enhance the quality standards of traditional Chinese medicine preparations.METHODS:UPLC-MS/MS was employed to identify the chemical constituents in Yuchuang powder.The chromatographic separation was performed on an ACQUITY UPLC HSS T3 column(100 mm×2.1 mm,1.8 μm)with gradient elution using a mobile phase consisting of 0.1%formic acid in water and acetonitrile.The injection volume was 10 μL.Automated screening of chemical components was conducted bu using UNIFI software based on the UNIFI traditional Chinese medicine database.The identified compounds were classified and verified by comparison with public databases and published literature,accompanied by fragment ion matching.RESULTS:A total of 80 chemical components were identified,of which 58 were detected in positive ion mode,41 in negative ion mode,and 19 components were detected in both ion modes.Divided by compound categories,it mainly includes 39 terpenes,7 phenanthrene compounds,8 phenolic acids,4 alkaloids,as well as flavonoids,vitamin D derivatives,fatty acids,and other components.CONCLUSIONS:The UPLC-MS/MS method established in the study can identify the pharmacologically active chemical components and characteristic constituents of each medicinal material in Yuchuang powder,providing reference for the rapid identification and quality control of the chemical components of the preparation.
