参芪地黄颗粒对局灶节段性肾小球硬化症大鼠足细胞损伤的保护作用及对HIF-1α、HMOX1、p27、PAI-1的影响OA
Protective effect of Shenqi Dihuang Granules on podocyte injury in focal segmental glomerulosclerosis rats and influence on HIF-1α,HMOX1,p27 and PAI-1
目的:明确参芪地黄颗粒对局灶节段性肾小球硬化症(FSGS)足细胞损伤的保护作用及其机制.方法:采用单侧肾切除联合阿霉素尾静脉注射构建大鼠FSGS模型,比色法检测24h尿蛋白定量(24hUTP)及血清白蛋白(ALB)、肌酐(Scr)、尿素氮(BUN)、总胆固醇(TC)、甘油三酯(TG)水平;PAS染色和电镜观察肾脏病理损伤情况;Western Blot检测差异蛋白的表达.结果:参芪地黄颗粒能够减轻FSGS大鼠肾脏病理损伤,降低24hUTP(P<0.01),升高血清ALB水平(P<0.05),降低Scr、BUN、TC、TG水平(P<0.01),升高足糖萼蛋白(PODXL)、肾小球裂隙膜蛋白(nephrin)、血红素加氧酶1(HMOX1)、p27蛋白表达水平(P<0.05),降低缺氧诱导因子-1α(HIF-1 α)、纤溶酶原激活物抑制剂1(PAI-1)蛋白表达水平(P<0.05).结论:参芪地黄颗粒能修复足细胞损伤,有效改善FSGS的肾脏病理,其机制与调控足细胞相关蛋白PODXL、nephrin及HIF-1 α通路相关分子HIF-1α、HMOX1、p27、PAI-1蛋白的异常表达有关.
Objective:To clarify the protective effect of Shenqi Dihuang Granules on focal segmental glomerulosclerosis(FSGS)podocyte injury,and to explore its underlying mechanism.Methods:Unilateral nephrectomy combined with adriamycin tail vein injection was used to construct a rat FSGS model,and 24-hour urinary protein quantification(24hUTP),serum plasma albumin(ALB),creatinine(Scr),urea nitrogen(BUN),total cholesterol(TC)and triglyceride(TG)levels were detected by colorimetry.PAS staining and electron microscopy were used to observe the pathological damage of the kidney.Western Blot was used to detect the expression of differentially expressed proteins.Results:Shenqi Dihuang Granules could alleviate the pathological damage of kidney,reduce 24hUTP(P<0.01),increase the ALB level(P<0.05),reduce the Scr,BUN,TC and TG levels(P<0.01),increase the protein expression levels of PODXL,nephrin,HMOX1 and p27(P<0.05),decrease the protein expression levels of HIF-1α and PAI-1(P<0.05).Conclusion:Shenqi Dihuang Granules can repair podocyte damage and effectively improve the renal pathology of FSGS,and its mechanism is related to the regulation of podocyte-related proteins PODXL,nephrin and HIF-1α pathway-related molecules HIF-1α,HMOX1,p27 and PAI-1 proteins.
