清金化痰颗粒的制备工艺及质量控制OA
Preparation process and quality control of Qingjin Huatan Granules
目的:优化清金化痰颗粒制备工艺,并对其进行质量控制.方法:采用层次分析法结合正交试验对清金化痰汤的水提工艺进行优化,进一步采用湿法制粒制备清金化痰颗粒,以成型率、吸湿率、休止角、制粒情况和溶化性为指标对制剂工艺进行评价和优选;同时结合薄层鉴别、HPLC含量测定和指纹图谱等方法建立清金化痰颗粒的质量标准,通过加速稳定性实验对清金化痰颗粒的稳定性进行初步研究.结果:清金化痰汤最佳提取工艺为10倍量水,提取2次,每次提取1 h;清金化痰颗粒最佳成型工艺以乳糖为辅料,药辅比1∶0.8,使用80%的乙醇润湿,制软材,制粒,60℃下干燥1 h,整粒,包装;建立了黄芩、栀子、橘红和知母4味药的薄层鉴别方法和黄芩苷、栀子苷、柚皮苷、芒果苷的含量测定方法;建立了 10批颗粒的指纹图谱,相似度为0.976~0.999,在指纹图谱中共标定18个共有峰,标定出其中9个色谱峰;且加速稳定性实验结果表明清金化痰颗粒稳定性良好.结论:该制备工艺及建立的指纹图谱方法稳定、可靠、重复性好,基于此建立了清金化痰颗粒的质量标准.
Objective:To optimize the preparation process of Qingjin Huatan Granules and to control its quality.Methods:The water extraction process of Qingjin Huatan Decoction was optimized by analytic hierarchy process combined with orthogonal experiment,and the preparation process of Qingjin Huatan Granules was further prepared by wet method.The formulation process was evaluated and optimized with the molding rate,moisture absorption rate,angle of repose,granulation performance and meltability as evaluation indicators.The quality standard of Qingjin Huatan Granules was established by thin-layer identification,HPLC content determination and fingerprint analysis.The stability of Qingjin Huatan Granules was studied by accelerated stability experiment.Results:The optimal extraction process of Qingjin Huatan Decoction was 10 times the volume of water,extract twice,and extract for 1 h each time.The optimal granulation process of Qingjin Huatan Granules was to use lactose as an excipient,with a drug-excipient ratio of 1∶0.8,wet with 80%ethanol,make soft material,granulate,dry at 60 ℃ for 1 h,whole grain,packaging.A thin-layer identification method was established for four medicinal herbs:Scutellaria baicalensis Georgi,Gardenia jasminoides Ellis,Citrus reticulata Blanco,and Anemarrhena asphodeloides Bge,as well as a content determination method for baicalin,geniposide,naringin and mangiferin.Ten batches of particle fingerprint patterns were established,with a similarity ranging from 0.976 to 0.999.A total of 18 common peaks were identified in the fingerprint patterns,and 9 chromatographic peaks were determined among them.The results of the accelerated stability test also show that the stability of Qingjin Huatan Granules is good.Conclusion:The preparation process and the established fingerprint chromatogram method are stable,reliable and have good repeatability.Based on this,the quality standard of Qingjin Huatan Granules has been established.
