鹿角胶通过调节性激素分泌与凋亡蛋白表达对延缓衰老的作用OA
Effects of anti-aging of Cervi Cornus Colla by regulating the secretion of sex hormones and the expression of apoptotic proteins
目的:研究鹿角胶通过调节性激素分泌以及凋亡蛋白表达对抗衰老的作用.方法:将2月龄大鼠设为青年对照组,18月龄雄性SD大鼠分为老年对照组、睾酮干预组和鹿角胶干预组,每组10只.睾酮干预组采用每周30 mg/kg睾酮干预,鹿角胶干预组采用0.6 g·kg-1·d-1鹿角胶干预.干预8周后,用ELISA检测血清中促性腺激素释放激素(GnRH)、黄体生成素(LH)、总睾酮(tT)和游离睾酮(fT)浓度,用Western Blot、IHC检测p16、p21、Caspase3、Ki67等凋亡相关蛋白水平.结果:青年对照组GnRH、LH、tT、fT均显著高于其余3组(P<0.05);老年对照组GnRH、LH水平低于睾酮干预组(P<0.05),tT、fT水平显著低于睾酮干预组和鹿角胶干预组(P<0.05).青年对照组p16、p21、Caspase3显著低于其余3组(P<0.05),老年对照组p16、p21、Caspase3显著过高于睾酮干预组和鹿角胶干预组(P<0.05).睾酮干预组Caspase3显著低于鹿角胶干预组(P<0.05);青年对照组Ki67显著高于其余3组(P<0.05),睾酮干预组Ki67显著高于老年对照组(P<0.05).结论:鹿角胶可能有通过调节性激素水平以及调控凋亡蛋白表达而抗衰老的作用.
Objective:To investigate the anti-aging effect of Cervi Cornus Colla through regulation of sex hormone secretion and apoptotic protein expression.Methods:Two-month-old rats were designated as the young control group(Group A).Eighteen-month-old male SD rats were randomly divided into three groups:aged control group(Group B),testosterone intervention group(Group C),and Cervi Cornus Colla intervention group(Group D),with 10 rats in each group.The testosterone group received testosterone at a dose of 30 mg/kg per week,while the Cervi Cornus Colla group received 0.6 g·kg-1·d-1 of Cervi Cornus Colla.After 8 weeks of intervention,serum concentrations of gonadotropin-releasing hormone(GnRH),luteinizing hormone(LH),total testosterone(tT),and free testosterone(fT)were measured by ELISA.The levels of apoptosis-related proteins,including p16,p21,Caspase-3,and Ki67,were detected by Western Blot and immunohistochemistry(IHC).Results:The levels of GnRH,LH,tT,and fT in the young control group were significantly higher than those in the other three groups(P<0.05).The aged control group showed lower GnRH and LH levels compared with the testosterone group(P<0.05),and significantly lower tT and fT levels than both the testosterone and Cervi Comus Colla groups(P<0.05).The young control group had significantly lower levels of p16,p21,and Caspase-3 than the other three groups(P<0.05).The aged control group exhibited significantly higher levels of p1 6,p21,and Caspase-3 compared with the testosterone and Cervi Comus Colla groups(P<0.05).The testosterone group had significantly lower Caspase-3 level than the Cervi Comus Colla group(P<0.05).The Ki67 level in the young control group was significantly higher than that in the other three groups(P<0.05),and the Ki67 level in the testosterone group was significantly higher than that in the aged control group(P<0.05).Conclusion:Cervi Comus Colla may exert anti-aging effects by regulating sex hormone levels and modulating apoptotic protein expression.
