严重急性呼吸综合征冠状病毒2(SARS-CoV-2)是新型冠状病毒病(COVID-19)的病原体,在过去数年中对全球公共卫生系统、社会经济和民众日常生活均造成了严重影响.SARS-CoV-2入侵宿主细胞是其生命周期和传播的关键环节,其入侵过程与多数包膜病毒类似,主要包括病毒附着、与宿主细胞受体结合以及内吞与膜融合步骤.鉴于该过程在病毒生命周期的重要地位,阐明SARS-CoV-2的入侵机制并开发靶向病毒入侵的抑制剂,已成为防治COVID-19的重要策略.本文综述了SARS-CoV-2入侵机制和靶向SARS-CoV-2入侵过程的抑制剂研究进展,旨在为防治SARS-CoV-2感染及开发应对其他包膜病毒威胁的新型抗病毒策略提供参考.

Severe Acute Respiratory Syndrome Coronavirus 2(SARS-CoV-2),the causative agent of COVID-19,has posed a severe challenge to global public health,socioeconomic stability,and daily lives over the past few years.Viral entry of SARS-CoV-2 into host cells is a critical step in its life cycle and transmission,and primarily involves viral attachment,binding to host cell receptors,and membrane fusion as with most of the enveloped viruses.Given the importance of this process in the viral life cycle,elucidating the mechanisms of viral entry and developing viral entry inhibitors have become important strategies for prevention and treatment of COVID-19.This review summarizes recent advances in inhib-itors targeting the entry process of SARS-CoV-2,and aims to provide a reference for developing novel antiviral strategies against SARS-CoV-2 infection and for countering the threats posed by other envel-oped viruses.
