加味当归芍药散对肺炎链球菌诱导幼鼠肾损伤的保护作用OA

Protective effect of Jiawei Danggui Shaoyao San against Streptococcus pneumoniae-induced renal injury in young rats

目的 采用反复肺炎链球菌(Streptococcus pneumoniae,S.pn)雾化感染的方式构建幼鼠感染相关性肾小球肾炎(infection-related glomerulonephritis,IRGN)动物模型,探究当归芍药散(Danggui Shaoyao San,DSS)及其加味组(加味麻黄、黄芪)对IRGN的肾脏保护作用.方法 3周龄SPF级雄性SD大鼠随机分组,采用雾化给菌、游泳联合给菌两种方式构建IRGN模型,给予DSS、加味DSS、麻黄-黄芪及奥司他韦灌胃干预.动态检测尿蛋白/肌酐比值,检测血清生化指标及肺泡灌洗液炎症因子,HE染色、透射电镜观察肺肾病理及肾组织超微结构,免疫组化检测MAC、IgM等沉积.结果给菌组及游泳联合给菌组大鼠均出现体重下降,肺肾病理损伤,炎症因子升高,尿蛋白/肌酐比值及血清生化指标异常,IgM沉积增多、足突特异性蛋白podocin下调、神经氨酸酶升高,且游泳联合给菌组损伤更为明显,提示模型构建成功并选择游泳联合给菌进行造模;DSS组、加味DSS组及麻黄-黄芪组均能改善肾损伤,调节尿蛋白/肌酐比值、血清生化等相关指标,其中加味DSS组在调节尿蛋白及TC、TG、ALB指标中效果最优.结论 成功建立符合临床特征的IRGN复合模型,证实DSS及其加味组对S.pn诱导的幼鼠IRGN具有保护作用,为相关病理机制研究及中医药防治提供实验依据.

Aim To explore the renal protective ef-fects of Danggui Shaoyao San(DSS)and its modified prescription(Herba Ephedrae and Radix Astragali)on an infection-related glomerulonephritis(IRGN)ani-mal model via establishing repeated nebulization of Streptococcus pneumoniae(S.pn)in immature rats.Methods SPF male SD rats at 3-week-old were ran-domly divided into two groups,and the IRGN model was established by two ways,namely,aerosol injec-tion and swimming injection,and then the rats were given intragastric administration of DSS,Jiawei DSS,Ephedra and Astragalus,and oseltamivir.Urinary pro-tein/creatinine ratio,serum biochemical indicators and inflammatory factors in bronchoalveolar lavage fluid were dynamically detected.HE staining and transmis-sion electron microscopy were used to observe the pa-thology of lung and kidney and the ultrastructure of renal tissue.MAC and IgM deposition were detected by immunohistochemistry.Results The weight of rats decreased,the pathological injury of lung and kidney,the increase of inflammatory factors,the abnormality of urinary protein/creatinine ratio and serum biochemi-cal indexes,the increase of IgM deposition,the de-crease of Podocin and the increase of neuraminidase were observed in rats of the bacteria group and the swimming combined bacteria group.The injury of the swimming combined bacteria group was more obvious,suggesting that the model was successfully constructed and the swimming combined bacteria group was se-lected;DSS group,Jiawei DSS group and Ephedra and Astragalus group could improve renal injury,regu-late urinary protein/creatinine ratio,serum biochemis-try and other related indicators,among which Jiawei DSS had the best effect.Conclusions The compound model of IRGN is successfully established,which con-firms that DSS and its modified prescriptions have pro-tective effects on IRGN induced by S.pn in young rats,and it provides experimental evidence for the study of related pathological mechanisms and the pre-vention and treatment of traditional Chinese medicine.
