[目的]了解云南省猪繁殖与呼吸综合征病毒(Porcine reproductive and respiratory syndrome virus,PRRSV)的流行情况和遗传变异特征,为云南省PRRSV的防控提供理论依据.[方法]2016-2024年,从云南16个州市采集13 803份样品,通过实时荧光定量PCR扩增ORF7基因以鉴定PRRSV阳性样品,采用全局与局部空间自相关分析,解析病毒的时空分布特征,通过RT-PCR扩增369份阳性样本的ORF5基因,对筛选获得的部分代表性毒株序列及从NCBI中获取的8条云南分离株ORF5基因序列进行遗传进化、序列相似性、编码氨基酸的变异特征和N-糖基化位点分析.[结果]2016-2024年云南省PRRSV阳性率为2.67%(369/13 803).全局空间自相关分析显示,云南省PRRSV阳性率随机分布;局部空间自相关分析显示,PRRSV显著聚集区域呈动态变化.基于ORF5基因的遗传进化分析表明,筛选获得的16株代表毒株及8株云南分离株均属于PRRSV-2型,聚类为谱系8(15株)、谱系1(4株)、谱系3(3株)和谱系5(2株);24株PRRSV毒株核苷酸序列相似性为80.5%~99.7%,氨基酸序列相似性为75.8%~99.5%.氨基酸突变分析表明,诱骗表位区在谱系1、3、5毒株呈V27A、L28P、V29A及N30S突变;中和抗原表位区在谱系1、3、8毒株呈H38Y、L39V/S/I突变;13个毒株具有R13和R151毒力位点.此外,谱系1在第30-32、32-34、33-35和57-59位氨基酸处增加了N-糖基化位点,第51-53位氨基酸处缺失NGT;谱系8在第33-35、34-36位氨基酸处增加了N-糖基化位点,第35-37和44-46位氨基酸缺失NSS和NLT.[结论]2016-2024年云南省PRRSV阳性率<10%,全局空间分布随机且局部聚集区动态变迁.流行毒株呈现多谱系共存,以谱系8为主要流行谱系.ORF5基因氨基酸位点和N-糖基化位点存在不同程度变异.必须加强持续性流行病学监测,并实施更具针对性的防控与预警措施.

[Objective]This study aimed to understand the prevalence and genetic variation characteristics of Porcine reproductive and respiratory syndrome virus(PRRSV)in Yunnan province,and provide a theoretical basis for the prevention and control of PRRSV in Yunnan province.[Method]From 2016 to 2024,13 803 samples were collected from 16 prefectures in Yunnan.ORF7 gene was amplified by Real-time quantitative PCR to identify PRRSV-positive samples.Global and local spatial autocorrelation analyses were employed to elucidate the spatiotemporal distribution characteristics of the virus.Subsequently,ORF5 gene of positive samples was amplified by RT-PCR.The ORF5 gene sequences from selected representative virus strains and 8 Yunnan isolates retrieved from NCBI,were subjected to analysis of genetic evolution,sequence similarity,variation characteristics of their encoded amino acids,and N-glycosylation sites.[Result]From 2016 to 2024,the positive rate of PRRSV in Yunnan province was 2.67%(369/13 803).Global spatial autocorrelation analysis indicated that the PRRSV positivity rate in Yunnan province was randomly distributed,while local spatial autocorrelation analysis revealed that the significant clustering areas exhibited dynamic changes.Genetic evolution analysis based on ORF5 gene showed that all 16 representative virus strains obtained in this study,and the 8 Yunnan isolates,were classified as PRRSV-2,clustering into Lineage 8(15 strains),Lineage 1(4 strains),Lineage 3(3 strains),and Lineage 5(2 strain).The nucleotide sequence identity of the 24 PRRSV strains ranged from 80.5%to 99.7%,and the amino acid sequence identity ranged from 75.8%to 99.5%.Amino acid mutation analysis indicated that the decoy epitope region had V27A,L28P,V29A,and N30S mutations in Lineage 1,3,and 5 strains.The neutralizing antigenic epitope region had H38Y and L39S/I mutations in Lineage 1,3,and 8 strains.7 strains possessed virulence sites R13 and R151.Additionally,Lineage 1 gained N-glycosylation sites at amino acid positions 30-32,32-34,33-35 and 57-59,with a loss of NGT at positions 51-53.Lineage 8 gained N-glycosylation sites at positions 33-35 and 34-36,with a loss NSS and NLT at positions 35-37 and 44-46.[Conclusion]From 2016 to 2024,the PRRSV positivity rate in Yunnan province was<10%.The global spatial distribution was random,while local clustering exhibited dynamic shifts over time.The prevalent virus strains exhibited co-circulation of multiple lineages,with Lineage 8 identified as the predominant epidemic lineage.Amino acid sites and N-glycosylation sites in the ORF5 gene exhibited varying degrees of variation.It was essential to strengthen continuous epidemiological surveillance and implement more targeted prevention,control,and early warning measures.
