梅花鹿产茸性状的全基因组关联分析OA
Genome-Wide Association Study of Antler Production Traits in Cervus nippon
[目的]本研究基于全基因组重测序(whole genome resequencing,WGR)技术对梅花鹿产茸性状进行全基因组关联分析(genome-wide association study,GWAS),以挖掘梅花鹿产茸性状的候选基因,为实现高产茸量个体的早期精准筛选与定向选育提供理论基础.[方法]采集 266 只产茸时期梅花鹿的血样并提取 DNA,通过DNBSEQ-T7 测序平台进行 WGR,使用 GEMMA 软件的混合线性模型对产茸性状进行 GWAS.利用ANNOVAR软件对关联的单核苷酸多态性(SNP)位点进行注释,进一步通过PCR扩增进行检测.[结果]梅花鹿血样 WGR 共获得 60 317 854个 SNPs位点,通过 GWAS共检测到 67个与产茸性状显著关联的 SNPs位点(P<1×10―6).结合基因功能注释结果,进一步选取3个SNPs位点,对其对应的基因(COMT1、FMNL2及CRYM)进行 PCR检测,结果显示,上述基因内部所含的与产茸性状关联的 SNP位点均存在多态性.[结论]本研究基于WGR 数据,通过 GWAS 共筛选获得 67 个与梅花鹿产茸性状潜在相关的 SNPs 位点,其中筛选到的 COMT1、FMNL2及CRYM可能是影响产茸性状的重要候选基因,这些候选基因可作为分子标记辅助育种的潜在靶点,为解析梅花鹿产茸性状的遗传基础提供了有效分子标记.
[Objective]This study conducted genome-wide association study(GWAS)based on whole genome resequencing(WGR)technology to identify candidate genes for antler production traits,providing a theoretical foundation for early precise selection and targeted breeding of high-antler-yield individuals.[Method]Blood samples were collected from 266 Cervus nippon during the antler growth period,and the genomic DNA was subsequently extracted.WGR was performed on the DNBSEQ-T7 platform and GWAS of antler production traits was conducted via the mixed linear model in GEMMA.The associated single nucleotide polymorphism(SNPs)were annotated using ANNOVAR,and further detection was performed via PCR amplification.[Result]A total of 60 317 854 SNPs were identified from Cervus nippon blood samples via WGR.Through GWAS,a total of 67 SNPs significantly associated with the antler-producing trait were identified(P<1×10―6).Based on the results of gene functional annotation,the COMT1,FMNL2,and CRYM genes corresponding to 3 SNPs were further selected for PCR detection,and the results demonstrated that all the SNPs associated with antler production traits within the aforementioned genes exhibited polymorphism.[Conclusion]Based on WGR data,this study identified 67 SNPs potentially associated with the antler production traits of Cervus nippon through GWAS.Among them,COMT1,FMNL2,and CRYM screened out might be important candidate genes affecting the antler production traits.These candidate genes could serve as potential targets for molecular marker-assisted breeding and provide effective molecular markers for analyzing the genetic basis of antler production traits in Cervus nippon.
