D-半乳糖诱导不同时间对衰老大鼠内皮细胞微粒及氧化应激的影响OA
Effects of D-galactose induction duration on endothelial cell microparticles and oxidative stress in aging rats
目的 探讨 D-半乳糖诱导不同时间对衰老大鼠内皮细胞微粒(EMP)及氧化应激水平的影响.方法 40 只 SD 大鼠随机分为空白 1 组、空白 2 组、模型 1 组、模型 2 组,分别给予 D-半乳糖诱导 6 周和12 周,制备衰老大鼠模型.采用 HE 染色观察大鼠胸主动脉及肝脏组织的形态变化,流式细胞术检测大鼠血浆中 EMP 的数量,ELISA 检测大鼠肝组织 SOD、MDA 的含量及血清中 IL-6的含量.结果 经 D-半乳糖诱导6 周后,与空白 1 组比较,大鼠血浆 CD54+EMP 水平显著升高(P<0.05),但血清 IL-6 含量仅有升高趋势(P>0.05),肝脏组织中 SOD 活性降低(P<0.05),MDA 含量也呈现一定的升高趋势(P>0.05),但差异并无统计学意义.HE 染色结果显示大鼠胸主动脉、肝脏组织的形态未出现明显衰老变化.而诱导 12 周后,模型 2 组大鼠血浆 CD54+EMP 水平明显升高(P<0.01),肝脏组织中 SOD 活性显著降低(P<0.01),MDA 含量显著升高(P<0.01),血清 IL-6含量显著升高(P<0.01),胸主动脉内膜明显增厚,内膜/中膜比值显著升高(P<0.05),同时肝细胞核的体积增大,核质深染,有较多空泡形成,出现明显的衰老表现.结论 用 D-半乳糖诱导 12 周可使大鼠出现明显的衰老变化,同时大鼠 EMP 数量及氧化应激水平随造模时间的延长而增加,而诱导 6 周不能使大鼠出现明显的衰老变化.
Objective To investigate the effects of D-galactose induction for different durations on endothelial cell microparticles(EMP)and oxidative stress levels in aging rats.Methods Forty Sprague-Dawley rats were randomly divided into four groups:blank group 1,blank group 2,model group 1 and model group 2.Rats in the model group were injected with D-galactose for 6 and 12 weeks,respectively,to establish an aging rat model.Morphological changes in the thoracic aorta and liver tissues were detected by hematoxylin/eosin(HE)staining.Plasma levels of EMP were measured by flow cytometry.Liver levels of superoxide dismutase(SOD)and malondialdehyde(MDA)and serum interleukin(IL)-6 were measured by enzyme-linked immunosorbent assay.Results After 6 weeks of modelling,plasma levels of CD54+EMP were increased significantly(P<0.05);SOD liver activity was decreased(P>0.05),and liver MDA and serum IL-6 levels were showed only an upward trend compared with the blank group 1(P>0.05);however,the differences were not significant(P>0.05).HE staining indicated no obvious aging characteristics in the thoracic aorta and liver tissue.After 12 weeks of modelling,however,plasma CD54+EMP levels increased significantly(P<0.01),SOD liver activity decreased(P<0.01),MDA levels were substantially elevated(P<0.01),and serum IL-6 levels were significantly higher compared with the 6-week modelling group(P<0.01).The thoracic aortic intima was noticeably thickened and the intima/media ratio was significantly increased(P<0.05).The hepatocyte nuclear volume was also markedly increased,nuclear and cytoplasmic staining deepened,and numerous vacuoles formed,showing obvious signs of aging.Conclusions Induction with D-galactose for 12 weeks can cause significant senescence changes in rats.Concurrently,EMP plasma levels and oxidative stress levels increased with increasing modelling period.In contrast,modelling for 6 weeks result ed in no significant aging characteristics in rats.
