基于荧光SSR标记的杧果杂交后代杂种真实性鉴定OA
Identification of mango hybrid progeny based on fluorescent SSR markers
杂交育种是杧果新品种选育最重要最有效的手段,快速高效鉴定杂交后代真实性是实现育种计划的前提.以"金煌(Mangifera indica L.'Jinhuang')"×"热农 1 号(M.indica L.'Renong No.1')"、"金煌"×"爱文(M.indica L.'Irwin')"、"热农1号"×"台农1号(M.indica L.'Tainong No.1')"杂交后代及亲本为试材,采用荧光SSR标记结合毛细管电泳技术筛选亲本间具有多态性的SSR引物,对其杂交后代单株真实性进行鉴定.结果表明,"金煌"×"热农1号"杂交F1的7个单株在7个位点上,"金煌"×"爱文"杂交F1的6个单株在6个位点上,"热农1号"×"台农1号"杂交F1的11个单株在4个位点上扩增出双亲的多态性条带,鉴定为真杂种.UPGMA聚类分析中,141132、141193与"金煌"聚成一类,1411100、130405和141166与"热农1号"聚成一类,而141182和141195分别单独为一类;"金煌"×"爱文"和"热农1号"×"台农1号"的F1单株均与父本聚为一类,表现出偏父本遗传的倾向.研究结果为利用荧光SSR分子标记进行杧果杂交种真实性鉴定提供重要参考.
Hybrid breeding is the most important and effective method for developing new mango varieties,and the rapid and efficient identification of hybrid progeny is a prerequisite for the implementation of breeding programs.In this study,the hybrid progeny and their parents from three crosses including'Jinhuang'×'Renong No.1','Jinhuang'×'Irwin',and'Renong No.1'×'Tainong No.1'were used as experimental materials.With capillary electrophoresis,fluorescent polymorphic SSR markers were screened between the parents and further used to identify their hybrid progeny.The results showed that seven in-dividuals of the'Jinhuang'×'Renong No.1'cross at seven loci,six individuals of the'Jinhuang'×'Irwin'cross at six lo-ci,and eleven individuals of the'Renong No.1'×'Tainong No.1'cross at four loci amplified polymorphic bands from both parents,indicating all of them were hybrids.In the UPGMA clustering analysis,141132 and 141193 were clustered with'Jin-huang',1411100,130405,and 141166 were clustered with'Renong No.1',while 141182 and 141195 were clustered in a sep-arate category.All F1 individuals from both'Jinhuang'×'Irwin'and'Renong No.1'×'Tainong No.1'crosses were clustered with their respective paternal parents,showing a tendency of paternal inheritance.The results of this study provide important theoretical basis and reference for the identification of mango hybrids using fluorescent SSR molecular markers.
