富民枳组培育苗及光合功能分析OA
Analysis of tissue culture and photosynthetic function of Poncirus polyandra
为保护和利用富民枳种质资源,以富民枳幼嫩茎段为外植体,进行离体再生体系建立研究,并对所得组培苗与实生苗的光合功能进行比较分析.结果表明,适宜腋芽诱导的最佳培养基为MT+6-BA 2.0 mg/L+IAA 0.2 mg/L+水解酪蛋白200 mg/L,诱导率达88.9%,且长势良好;适宜不定芽增殖的最佳培养基为WPM+6-BA 0.5 mg/L+IAA 0.5 mg/L+粉状活性炭0.2 g/L,增殖系数为8.7;适宜生根诱导的培养基为MT+6-BA 1.0 mg/L+IBA 0.5 mg/L+粉状活性炭0.2 g/L,生根率为73.33%,生根系数为5.85.组培苗叶片蒸腾速率显著高于实生苗,且叶绿素含量也显著高于实生苗.富民枳组培苗光合功能优于实生苗,但对干旱的耐受能力较弱,应注意加强田间抗旱管理.
To protect and utilize the germplasm resources of Poncirus polyandra,its young stem segments were used as explants to establish an in vitro regeneration system,and the photosynthetic function of the tissue-cultured seedlings was com-pared with that of seed-grown seedlings.The results showed that the optimal medium for axillary bud induction was MT+6-BA 2.0 mg/L+IAA 0.2 mg/L+casein hydrolysate(CH)200 mg/L,with an induction rate of 88.9%and vigorous growth.The optimal medium for adventitious bud proliferation was WPM+6-BA 0.5 mg/L+IAA 0.5 mg/L+activated carbon(AC)0.2 g/L,with a proliferation coefficient of 8.7.The optimal medium for root induction was MT+6-BA 1.0 mg/L+IBA 0.5 mg/L+powdered activated carbon(PAC)0.2 g/L,with a rooting rate of 73.33%and a rooting coefficient of 5.85.The transpiration rate and chlorophyll content of tissue-cultured seedling leaves were both significantly higher than those of the seed-grown seedlings.The photosynthetic function of tissue-cultured seedlings was superior to that of seed-grown seedlings,but their tolerance to drought was reduced.Therefore,attention should be paid to strengthening field drought-resistance management.
