过氧亚硝酸盐(ONOO-)在细胞信号转导及体内平衡调节中发挥着重要作用,实现ONOO-的检测对疾病的早期诊断和治疗至关重要.本文以罗丹明B和对氨基苯酚为前体,采用一步溶剂热法成功制备了橙色荧光碳点(O-CDs).所制备的O-CDs因表面的氨基/羟基官能团可被ONOO-氧化,对其表现出良好的选择性和荧光猝灭响应.当ONOO-浓度处于30~480和670~1070 nmol/L范围时,其浓度与荧光强度变化的线性相关性良好,检出限为9 nmol/L.与此同时,随着ONOO-浓度变化,溶液颜色由粉红色逐渐变为深蓝色,吸光度与ONOO-浓度在300~1000 nmol/L范围内呈良好线性响应,检出限为90 nmol/L.因此,该工作成功实现了对ONOO-的荧光和比色双模式的检测.此外,凭借其优异的光稳定性及低毒性等特性,O-CDs成功应用于PC-12活细胞中ONOO-的荧光成像,这为阐释ONOO-在相关病理过程中复杂的生物学作用提供了重要手段.

Peroxynitrite(ONOO-)plays an important role in cellular signal transduction and homeostasis;thereby,achieving the detection of ONOO-is crucial for early diagnosis and treatment of diseases.Herein,orange fluorescent carbon dots(O-CDs)were successfully synthesized by a one-step solvothermal method using rhodamine B and p-aminophenol as precursors.The O-CDs exhibited good selectivity and a sensitive response to ONOO-due to the oxidation of the amino/hydroxyl functional groups on their surface by ONOO-.Within the concentration ranges of 30~480 and 670~1070 nmol/L,the quenching efficiency of O-CDs showed a good linear correlation with the concentration of ONOO-,with the detection limit of 9 nmol/L.Meanwhile,the color of the solution gradually changed from pink red to dark blue.For the colorimetric mode,a good linear relationship between the absorbance change and ONOO-concentration was obtained in the range of 300~1000 nmol/L,with the detection limit of 90 nmol/L.Consequently,this work has achieved the fluorescent and colorimetric dual-mode specific recognition and detection of ONOO-.In addition,due to the excellent photostability and low toxicity of O-CDs,they have been successfully applied for the fluorescent imaging of ONOO-in PC-12 living cells,providing a valuable tool for elucidating the complex biological roles of ONOO-in related pathological processes.
