杏黄止咳合剂通过PI3K/Akt/NF-κB信号通路治疗哮喘小鼠气道炎症机制研究OA
Effect and Mechanism of Xinghuang Zhike Mixture on Reducing Airway Inflammation in Asthmatic Mice Through PI3K/Akt/NF-κB Signaling Pathway
目的:探讨杏黄止咳合剂对小鼠哮喘气道炎症的保护作用及机制.方法:将小鼠随机分为正常对照组、模型组、杏黄止咳合剂组、杏黄止咳合剂联合布地奈德组.采用卵清蛋白致敏、激发诱导建立小鼠哮喘模型.除正常对照组外,模型组、杏黄止咳合剂组、杏黄止咳合剂联合布地奈德组于第1、7、14天腹腔注射卵清蛋白和氢氧化铝致敏.第21天开始,用卵清蛋白溶液雾化激发,连续激发14d,每次30min.杏黄止咳合剂组于激发前1h灌胃杏黄止咳合剂(0.13mL/10g),1次/d,连续4周;杏黄止咳合剂联合布地奈德组于激发前1h灌胃杏黄止咳合剂(0.13mL/10g),并雾化吸入0.01%布地奈德2mL 20min,1次/d,连续给药4周;正常对照组与模型组给予0.9%生理盐水进行同等操作.末次给药24h后麻醉处死小鼠,收集小鼠血清及肺泡灌洗液进行细胞分类和计数,HE染色观察小鼠肺组织病理形态,ELISA检测小鼠肺泡灌洗液中白细胞介素-5(IL-5)、白细胞介素-13(IL-13)和血清中白细胞介素-4(IL-4)、免疫球蛋白E(IgE)的 水 平,Western Blot检测肺组织中p-PI3K、PI3K、p-Akt、Akt、NF-κB、p56蛋白表达水平.结果:杏黄止咳合剂组、杏黄止咳合剂联合布地奈德组能显著降低哮喘小鼠肺泡灌洗液中白细胞数量(P<0.001),对哮喘小鼠肺组织病理损伤有改善作用,可明显降低小鼠肺泡灌洗液中IL-5、IL-13和血清中IL-4、IgE炎症因子水平(P<0.01),对哮喘模型小鼠p-PI3K/PI3K、p-Akt/Akt、NF-κB、p56蛋白表达有显著回调作用(P<0.01).结论:杏黄止咳合剂可能通过抑制PI3K/Akt/NF-κB信号通路的表达改善哮喘小鼠气道炎症,从而保护肺组织.
Objective:To investigate the protective effect and mechanism of Xinghuang Zhike Mixture on airway inflam-mation in asthmatic mice.Methods:Mice were randomly divided into normal control group,model group,Xinghuang Zhike Mixture group and Xinghuang Zhike Mixture combined with budesonide group.Asthmatic mice model was estab-lished by ovalbumin sensitization and stimulation.In addition to the normal control group,the model group,Xinghuang Zhike Mixture group and Xinghuang Zhike Mixture combined with budesonide group were sensitized by intrabitoneal injection of ovalbumin and alumina hydrogen on the first,seventh and fourteenth day.From day twenty-one,mice were atomized and stimulated with ovalbumin solution continuously for fourteen days for 30 minutes each time.Xinghuang Zhike Mixture group was given intragastric administrationof Xinghuang Zhike Mixture(0.13mL/10g)one hour before stimulation,once a day for four weeks.Xinghuang Zhike Mixture combined with budesonide group was given intragas-tric administration of Xinghuang Zhike Mixture(0.13mL/10g)one hour before stimulation,and atomized inhalation of 0.01%budesonide 2mL for twenty minutes,once a day for four weeks.Normal control group and model group were given intragastric administration of 0.9%normal saline once a day for four weeks.The mice were anesthetized twenty four hours after last administration.The serum and alveolar lavage fluid of mice were collected for cell classification and counting.The pathological morphology of lung tissue was observed by hematoxylin-eosin staining.The contents of in-terleukin 5 and Interleukin-13 in alveolar lavage fluid and Interleukin-4 and Immunoglobulin E in serum were detected by Enzyme linked immunosorbent assay.Western Blot analysis protein expression levels in lung tissue of p-PI3K,PI3K,p-Akt,Akt,NF-κB、p56.Results:Xinghuang Zhike Mixture group and Xinghuang Zhike Mixture combined with budesonide group can significantly reduce(P<0.001)the number of white blood cells in the alveolar lavage fluid of asthmatic mice,pathological injury of lung tissue of asthmatic mice also improved.The levels of interleukin 5,interleu-kin-13,Interleukin-4 and Immunoglobulin Ereduced significantly(P<0.01).Xinghuang Zhike Mixture can significantly regulate(P<0.01)the expression of p-PI3K/PI3K,p-Akt/Akt,NF-κB p65 in asthmatic mice.Conclusion:Xinghuang Zhike Mixture may improve airway inflammation of asthmatic mice by inhibiting the expression of PI3K/Akt/NF-κB signaling pathway to protect lung tissue of mice.
