Study on Extraction and Purification Process of Corn Glycogen and Determination of Its Moisture Retention and Hygroscopicity

以新鲜甜玉米为原料,采用机械破碎-溶剂沉淀法制备玉米糖原粗提物,通过考马斯亮蓝 G-250 法展开蛋白质含量的测定,进而比较调节 pH 法、煮沸法及两者联用法的脱蛋白效果,最终测定纯化产物的保湿与吸湿性能.结果表明:3 种脱蛋白方法中,调节 pH 4.9 结合煮沸30 min 的联用法效果最优,可将玉米糖原中蛋白质质量分数降至6.44%,显著低于调节 pH 法(最低 8.02%)与煮沸法(最低 9.50%);25℃条件下,在相对湿度41%的环境中,纯化后玉米糖原的保湿性能显著优于海藻酸钠与丙三醇,而其吸湿性能明显弱于上述两种对照物质.

Fresh sweet corn was used as the raw material to prepare crude corn glycogen extract via the mechanical crushing-solvent precipitation method.Protein content was determined based on the Coomassie Brilliant Blue G-250 method,Then the deproteinization effects of pH adjust-ment,boiling,and the combined method of pH adjustment and boiling were compared.Finally the moisturizing and hygroscopic properties of the purified product were determined.The results showed that among the three deproteinization methods,the combined method(adjusting pH to 4.9 and boiling for 30 min)showed the best effect,reducing the protein content in corn glycogen to 6.44%,which was significantly lower than that of the pH adjustment method(minimum 8.02%)and the boiling method(minimum 9.50%).Under the conditions of 25℃and 41%relative humidity,the moisturizing performance of the purified corn glycogen was significantly superior to that of sodium alginate and glycerol,while its hygroscopic performance was obviously weaker than that of the two aforementioned substances.
