圆口铜鱼养殖群体和野生群体遗传多样性比较分析OA

[目的]通过比较圆口铜鱼养殖群体和野生群体的遗传多样性水平,为圆口铜鱼人工繁育和增殖放流活动提供理论依据和技术参考.[方法]利用微卫星分子标记,对采集的1个野生群体和2个养殖群体共100尾圆口铜鱼样本进行种群遗传学分析.[结果]15个微卫星标记共检测出205个等位基因,3个圆口铜鱼群体均表现出较高的遗传多样性(Ho=0.630~0.742,He=0.687~0.787).遗传分化和分子方差分析结果表明,3个群体中养殖群体A1和A2属于中等程度分化(0.05<Fst<0.15),其余群体间的遗传分化偏低;圆口铜鱼有92.89%的遗传变异来源于群体内,个体变异是遗传变异的主要来源.UPGMA进化树聚类结果指示野生群体(W)和养殖群体(A1)的亲缘关系最近,2个养殖群体之间的亲缘关系最远,与PCoA主坐标分析结果一致.[结论]圆口铜鱼野生群体遗传多样性高于养殖群体,建议增加繁殖亲本的数量,选择遗传距离较远的亲本进行人工繁育,提高放流苗种的遗传多样性,避免持续的增殖放流活动改变圆口铜鱼的遗传变异组成.

[Objective]By comparing the differences in genetic diversity between cultured and wild populations of Coreius guichenoti,this study aimed to provide a theoretical basis and technical reference for the artificial breeding and release activities.[Method]A total of 100 samples were collected from one wild population and two cultured populations.Population genetic analysis was conducted using microsatellite molecular markers.[Result]A total of 205 alleles were detected across 15 microsatellite markers.All three populations of C.guichenoti exhib-ited high genetic diversity(Ho=0.630-0.742,He=0.687-0.787).Genetic differentiation and molecular analysis of variance revealed that the cultured populations A1 and A2 were moderately differentiated(0.05<Fst<0.15);genetic differentiation among the remaining populations was low.Approximately 92.89%of the genetic variation in C.guichenoti originated within populations,indicating that individual variation was the primary source of genetic diversity.Clustering analysis based on the UPGMA method indicated that the wild population(W)was the most closely related to cultured population(A1),while the two cultured populations were the most genetically distant.This finding was in agreement with the principal coordinate analysis(PCoA).[Conclusion]The wild population of C.guichenoti exhibite higher genetic diversity than the cultured populations.It is recommended to update the broodstocks and select individuals with greater genetic distance for artificial breeding to improve the genetic diversity of released juveniles.Additionally,continuous release activities should be avoided to pre-vent alterations in the genetic composition of C.guichenoti.
