[目的]探究凌云牛心李叶绿体基因组结构特征及系统发育关系,为其分子鉴定、遗传多样性研究及种质资源保护与开发利用提供理论依据.[方法]采用Illumina NovaSeq测序平台对凌云牛心李叶绿体基因组进行测序,利用生物信息学软件进行序列拼接、注释并对其结构特征、密码子偏好性、简单重复序列(SSR)和系统发育关系进行分析.[结果]凌云牛心李叶绿体基因组为典型的环状四分体结构,全长为157 926 bp,2个反向重复区(IR)长度均为26 353 bp,大单拷贝区(LSC)和小单拷贝区(SSC)长度分别为86 189和19 031 bp,叶绿体基因组总GC含量为37.53%.凌云牛心李叶绿体基因组共有130个基因,包括85个蛋白编码基因、37个tRNA基因和8个rRNA基因.对凌云牛心李叶绿体基因组蛋白编码基因的同义密码子相对使用度(RSCU)进行分析,结果表明高频密码子(RSCU>1)有32种,其中以A或U结尾的高频密码子占84.38%.凌云牛心李叶绿体基因组共检测到73个SSR位点,包含单核苷酸重复、二核苷酸重复、四核苷酸重复和五核苷酸重复,其中单核苷酸重复基序以A/T的重复次数最多.将凌云牛心李与其他5种李属植物的叶绿体基因组IR边界进行比较,发现其IR边界相对保守.系统发育分析结果表明,凌云牛心李与蜂糖李亲缘关系最近,推测其属于中国李类型.[结论]凌云牛心李叶绿体基因组高度保守,密码子偏好以A或U结尾,系统发育分析结果表明凌云牛心李可能属于中国李类型.

[Objective]The paper aimed to study the structural characteristics and phylogenetic relationships of the chloroplast genome of Lingyun Niuxinli,and provide theoretical basis for its molecular identification,genetic diversity research,and germplasm resource protection and development.[Method]Illumina NovaSeq sequencing platform was used to sequence the chloroplast genome of Lingyun Niuxinli.Bioin-formatics software was used for sequence splicing,annotation,and analysis of its structural features,codon preferences,simple repeat se-quences(SSR),and phylogenetics.[Result]The chloroplast genome of Lingyun Niuxinli was a typical circular tetrad structure,with a total length of 157 926 bp,and two reverse repeat regions(IR)of 26 353 bp.The length of the large single copy region(LSC)and the small sin-gle copy region(SSC)were 86 189 bp and 19 031 bp,respectively.The total GC content of the chloroplast genome was 37.53%.The chloro-plast genome of Lingyun Niuxinli contained a total of 130 genes,including 85 protein coding genes,37 tRNA genes,and 8 rRNA genes.The relative usage of synonymous codons(RSCU)for the chloroplast genome protein coding genes of Lingyun Niuxinli was analyzed,and the re-sults showed that there were 32 high-frequency codons(RSCU>1),of which the high-frequency codons ending in A or U accounted for 84.38%.A total of 73 SSR loci were detected in the chloroplast genome of Lingyun Niuxin plum,including single nucleotide repeats,dinucleoti-de repeats,tetranucleotide repeats,and pentanucleotide repeats.Among them,the A/T repeat sequence had the highest number of single nu-cleotide repeat motifs.Comparing the chloroplast genome IR boundaries of Lingyun Niuxin plum with five other plum species,it was found that its IR boundaries were relatively conserved.The phylogenetic analysis results indicated that Lingyun Niuxin plum had the closest genetic relationship with Fengtang plum,suggesting that it belonged to the Chinese plum type.[Conclusion]The chloroplast genome of Lingyun Niux-in plum is highly conserved,with codon preferences ending in A or U.Phylogenetic analysis suggests that Lingyun Niuxin plum may belong to the Chinese plum type.
