活筋壮骨散对大鼠骨关节炎的治疗作用研究OA

Study on the therapeutic effect of Chinese herbal compound HJZGS on osteoarthritis in rats

旨在借助骨关节炎(osteoarthritis,OA)大鼠模型评估活筋壮骨散(Huo Jin Zhuang Gu San,HJZGS)对 OA 的治疗效果,探究其潜在作用机制.在关节腔内局部注射碘乙酸钠建立 OA 大鼠模型,HJZGS 灌胃治疗21 d 后,通过组织学观察和检测血清细胞因子水平评估治疗效果.使用脂多糖(LPS)在 RAW 264.7 细胞中诱导炎症反应,检测 HJZGS 干预后的巨噬细胞极化、线粒体膜电位、活性氧(ROS)产生、细胞周期和凋亡情况,来评估 HJZGS 在体外的抗炎能力.结果:体内研究显示,HJZGS 能够显著减轻 OA 大鼠的关节软骨损伤(包括软骨细胞坏死及软骨基质破坏),同时降低血清中的促炎细胞因子白介素-6(IL-6)、肿瘤坏死因子 α(TNF-α)等的水平.体外研究显示,HJZGS 能够调节巨噬细胞极化,促进巨噬细胞由 M1 向 M2 表型极化,挽救线粒体膜电位,减少 ROS 的产生,逆转 LPS 导致的细胞周期阻滞并减少细胞凋亡,从而最大限度地减少细胞损伤.综上,HJZGS 能通过调节巨噬细胞极化,抑制炎症和氧化应激,有效缓解碘乙酸钠诱导的大鼠骨关节炎,这一发现为HJZGS 在兽医临床中治疗 OA 提供了试验依据,同时也为开发功能性宠物食品添加剂奠定了理论基础.

Osteoarthritis(OA)is a common chronic disease,characterized mainly by cartilage degeneration,joint pain,inflammation,and gradual loss of joint function.Huojin Zhuanggu San(HJZGS),a traditional Chinese medicine compound,composed of six herbal ingredi-ents,was evaluated in this study for its therapeutic effects and potential mechanisms of action on OA using an OA rat model.In vivo,the OA rat model was established by intra-articular injection of sodium monoiodoacetate.After 21 days of oral administration of HJZGS,the thera-peutic effects were assessed by histological observation and measurement of serum cytokine levels.In vitro,inflammation was induced in RAW 264.7 cells using LPS.The polarization of macrophages,mitochondrial membrane potential,reactive oxygen species(ROS)produc-tion,cell cycle and apoptosis were detected after HJZGS intervention to evaluate the anti-inflammatory ability of HJZGS.The in vivo study demonstrated that HJZGS significantly alleviated joint cartilage damage in OA rats and reduced the levels of pro-inflammatory cytokines such as IL-6 and TNF-α in serum.The in vitro study showed that HJZGS modulated macrophages polarization,promoting the shift of macrophages from the M1 to the M2 phenotype,thereby exerting anti-inflammatory effects.Additionally,HJZGS reduced the production of ROS,en-hanced cellular antioxidant capacity,and rescued mitochondrial membrane potential,thereby minimizing cell damage.HJZGS effectively alle-viated sodium monoiodoacetate-induced OA in rats by modulating macrophages polarization and inhibiting inflammation and oxidative stress.This finding provided experimental evidence for application of HJZGS in veterinary treatment of canine OA and laid a theoretical foundation for development of functional pet food additives.
