枸橘-壁虎通过调控PI3K/Akt通路对三阴性乳腺癌细胞增殖、凋亡和上皮间质转化的影响OA
Poncirus trifoliata with Gecko inhibits proliferation,apoptosis and epithelial-mesenchymal transition of triple negative breast cancer cells via regulating the PI3K-Akt signaling pathway
目的 探讨枸橘-壁虎对三阴性乳腺癌4T1 细胞增殖、凋亡和上皮间质转化(EMT)的影响及其作用机制.方法 取4T1 细胞,CCK-8 法检测不同浓度枸橘-壁虎干预后细胞存活率,确定后续实验枸橘-壁虎浓度.然后设空白组及枸橘-壁虎低、中、高剂量组,划痕实验检测细胞迁移能力(计算细胞划痕愈合率),流式细胞仪检测细胞凋亡率,qPCR 法检测细胞中 EMT 相关标志物神经型钙黏蛋白(N-cadherin)、波形蛋白(Vimentin)、上皮型钙黏蛋白(E-cadherin)及磷脂酰肌醇3-激酶(PI3K)、蛋白激酶 B(Akt)mRNA 表达情况,Western blot 法检测细胞中 N-cad-herin、Vimentin、E-cadherin 及 PI3K、p-Akt、Akt(此 3 项检测增设枸橘-壁虎高剂量+SC79 组)蛋白表达情况.结果 培养24 h 时枸橘-壁虎干预4T1 细胞的IC50 值为8.069 mg/mL,确定4,6,8 mg/mL 为枸橘-壁虎低、中、高剂量进行后续研究.与空白组比较,枸橘-壁虎各剂量组细胞划痕愈合率及细胞中 N-cadherin、Vimentin、Akt mRNA 相对表达量和 N-cadherin、Vimentin、PI3K、p-Akt、Akt 蛋白相对表达量均明显降低(P 均<0.05),枸橘-壁虎各剂量组细胞凋亡率、细胞中 E-cadherin mRNA 相对表达量和枸橘-壁虎中、高剂量组细胞中 E-cadherin 蛋白相对表达量均明显升高(P 均<0.05),且枸橘-壁虎高剂量组各指标(除 PI3K mRNA 相对表达量)较枸橘-壁虎低、中剂量组改善明显(P 均<0.05).枸橘-壁虎高剂量+SC79 组 PI3K、p-Akt、Akt 蛋白相对表达量均明显高于枸橘-壁虎高剂量组(P 均<0.05).结论 枸橘-壁虎可抑制4T1 细胞增殖、迁移和 EMT,促进细胞凋亡,其作用机制可能与抑制 PI3K/Akt 通路有关.
Objective It is to investigate the effects and mechanism of Poncirus trifoliata with Gecko on the prolifera-tion,apoptosis and epithelial-mesenchymal transition(EMT)of triple negative breast cancer(TNBC)4T1cells.Methods 4T1 cells were taken and cultured,and their cell viabilities after treatment with different concentrations of Poncirus trifoliata with Gecko were assesed by CCK-8 assay to determine the appropriate concentration for subsequent experiments.A blank group and low-dose,medium-dose,and high-dose groups of Poncirus trifoliata with Gecko were set up in the experiment.The cell migration ability was assessed by scratch assay(cell scratch healing rate was calculated),the cell apoptosis rate was determined by flow cytometry,the mRNA expressions of EMT-related markers in cells,including N-cadherin,vimentin and E-cadherin,as well as phosphoinositide 3-kinase(PI3K)and protein kinase B(Akt)were detected by and qPCR,the pro-tein expressions of N-cadherin,vimentin,E-cadherin,as well as PI3K,p-Akt and Akt(these three indicators were addition-ally detected in the high-dose Poncirus trifoliata with Gecko+SC79 group)were detected by Western blot analysis.Results After 24 hours of culture,the IC50 value of Poncirus trifoliata with Gecko in the treatment of 4T1 cells was 8.069 mg/mL,4,6,and 8 mg/mL were selected as the low,medium,and high doses of Poncirus trifoliata with Gecko for the subsequent studies.Compared with the blank group,the cell scratch healing rate and the relative mRNA expressions of N-cadherin,vi-mentin,and Akt,as well as the relative protein expressions of N-cadherin,vimentin,PI3K,p-Akt and Akt in each dose groups of Poncirus trifoliata with Gecko were significantly reduced(all P<0.05),while the cell apoptosis rate,relative mRNA expression of E-cadherin in the cells of each does groups of Poncirus trifoliata with Gecko,and the relative protein expressions of E-cadherin in the cells of the medium-dose and high-dose groups of Poncirus trifoliata with Gecko were signif-icantly increased(all P<0.05),and the improvements of all indicators(except for the relative mRNA expression of PI3K)in the high-dose group of Poncirus trifoliata with Gecko were more significant than those in the low-dose and medi-um-dose groups(all P<0.05).The relative protein expressions of PI3K,p-Akt and Akt in the high-dose Poncirus trifolia-ta with Gecko+SC79 group were all significantly higher than those in the high-dose group of Poncirus trifoliata with Gecko(all P<0.05).Conclusion Poncirus trifoliata with Gecko can inhibit the proliferation,migration and EMT of 4T1 cells,and promote their apoptosis,the mechanism may be related to the inhibition of PI3K-Akt pathway.
