目的 基于 NOD 样受体热蛋白结构域相关蛋白3(NLRP3)/半胱氨酸天冬氨酸蛋白酶-1(Caspase-1)/焦孔素 D(GSDMD)信号通路探讨健脾消癌方对大肠癌细胞焦亡的影响.方法 将健脾消癌方煎煮浓缩至1.00 g/mL,采用含10%胎牛血清的PRMI1640 培养液调整为含生药量0.00 g/mL、0.25 g/mL、0.50 g/mL、1.00 g/mL 的含药血清;同时取人 NLRP3 重组蛋白(rhNLRP3),采用含10%胎牛血清的 PRMI1640 培养液调整为含药量10 μmol/mL 的含药血清.以人大肠癌 HCT116 细胞进行研究,实验设8 组,分别为汤药0.00 g/mL 组、汤药0.25 g/mL 组、汤药0.50 g/mL 组、汤药1.00g/mL 组、rhNLRP3 10 μmol/mL 组、汤药0.25 g/mL+rhNLRP3 10 μmol/mL组、汤药0.50 g/mL+rhNLRP3 10 μmol/mL 组、汤药1.00 g/mL+rhNLRP3 10 μmol/mL 组.CCK-8 法检测细胞的增殖率,平板克隆形成实验检测细胞的克隆形成能力,免疫荧光化学法检测细胞中剪切型 Caspase-1(Cleaved Caspase-1)和 GSDMD 阳性表达情况,ELISA 法检测细胞培养上清液中白细胞介素(IL)-18 和 IL-1β 水平.结果 与汤药0.00 g/mL 组比较,汤药0.25 g/mL 组、汤药0.50g/mL 组、汤药1.00g/mL 组、rhNLRP3 10 μmol/mL 组、汤药0.25 g/mL+rhNLRP3 10 μmol/mL组、汤药0.50 g/mL+rhNLRP3 10 μmol/mL 组、汤药1.00 g/mL+rhNLRP3 10 μmol/mL 组细胞增殖率和细胞中 GSDMD 阳性表达平均光密度值均明显降低(P 均<0.05),细胞克隆数目均明显减少(P 均<0.05),细胞中 Cleaved Caspase-1 阳性表达平均光密度值和 IL-1β、IL-18 水平均明显升高(P 均<0.05).单纯汤药各浓度组细胞增殖率和细胞中 GSDMD 阳性表达平均光密度值均随汤药浓度升高而降低,细胞克隆数目均随汤药浓度升高而减少,细胞中 Cleaved Caspase-1 阳性表达平均光密度值和 IL-1β、IL-18 水平均随汤药浓度升高而降低,两两比较差异均有统计学意义(P均<0.05).与 rhNLRP3 10 μmol/mL 组比较,汤药+rhNLRP3 各组细胞增殖率和细胞中 GSDMD阳性表达平均光密度值均明显降低(P 均<0.05),细胞克隆数目均明显减少(P 均<0.05),细胞中 Cleaved Caspase-1 阳性表达平均光密度值和 IL-1β、IL-18 水平均明显升高(P 均<0.05);且汤药+rhNLRP3 各组细胞增殖率、细胞克隆数目、细胞中 GSDMD 和 Cleaved Caspase-1 阳性表达平均光密度值、细胞中 IL-1β 和 IL-18 水平均随汤药浓度升高而升高或降低,两两比较差异均有统计学意义(P 均<0.05).结论 健脾消癌方可抑制大肠癌 HCT116 细胞增殖,并可通过激活NLRP3/Caspase-1/GSDMD 信号通路,释放大量 IL-1β、IL-18,促进癌细胞焦亡.

Objective It is to investigate the effect of prescription for fortifying spleen and dispersing cancer on the py-roptosis of colorectal cancer cells through the regulation of the NOD-like receptor thermal protein domain associated protein 3(NLRP3)/Caspase-1/gasdermin D(GSDMD)signaling pathway.Methods The prescription for fortifying spleen and dispersing cancer was decocted and concentrated to 1.00 g/mL,then diluted with PRMI1640 medium containing 10%fetal bovine serum to prepare drug-containing serum solutions with drug concentrations of 0.00 g/mL,0.25 g/mL,0.50 g/mL,and1.00 g/mL;Concurrently,recombinant human NLRP3(rhNLRP3)was taken and diluted with PRMI1640 medium containing 10%fetal bovine serum to prepare drug-containing serum solution with a concentration of 10 μmol/mL.Human colorectal cancer HCT116 cells were used as the research subjects and divided into8 groups:which were0.00 g/mL herbal decoction group,0.25 g/mL herbal decoction group,0.50 g/mL herbal decoction group,1.00 g/mL herbal decoction group,10 μmol/mL rhNLRP3 group,0.25 g/mL herbal decoction+10 μmol/mL rhNLRP3 group,0.50 g/mL herbal de-coction+10 μmol/mL rhNLRP3 group,1.00 g/mL herbal decoction+10 μmol/mL rhNLRP3 group.The cell proliferation rate of each group was detected by CCK-8 method,the colony formation ability of the cells was assessed by colony formation assay,the positive protein expressions of cleaved-Caspase1 and GSDMD were detected by immunofluorescence chemistry,and the levels of interleukin-18(IL-18)and interleukin-1β(IL-1β)in cell culture supernatant were measured by ELISA.Results Compared with the0.00 g/mL herbal decoction group,the cell proliferation rates and mean optical density values of GSDMD positive expression were significantly decreased in the 0.25 g/mL herbal decoction group,0.50 g/mL herbal decoc-tion group,1.00 g/mL herbal decoction group,10 μmol/mL rhNLRP3 group,0.25 g/mL herbal decoction+10 μmol/mL rhNLRP3 group,0.50 g/mL herbal decoction+10 μmol/mL rhNLRP3 group,1.00 g/mL herbal decoction+10 μmol/mL rhNLRP3 group(all P<0.05),the numbers of cell colonies were significantly reduced(all P<0.05),and the mean op-tical density values of cleaved Caspase-1 positive expression,as well as the levels of IL-1β and IL-18 in the cells were sig-nificantly increased(all P<0.05).The cell proliferation rate and the mean optical density of GSDMD positive expression in cells were decreased as the decoction concentration increased in each simple herbal decoction group,while the numbers of cell colonies were also decreased as the decoction concentration increased,the mean optical density values of cleaved Caspase-1 positive expression and the levels of IL-1β and IL-18 in the cells were all decreased as the concentration of the herbal decoction increased,all pairwise comparisons showed statistically significant differences(all P<0.05).Compared with the 10 μmol/mL rhNLRP3 group,the cell proliferation rates and mean optical density values of GSDMD positive ex-pression in the cells were obviously decreased,and the numbers of cell colonies were significantly reduced in each herbal decoction+rhNLRP3 group(all P<0.05),while the mean optical density values of cleaved Caspase-1 positive expression and the levels of IL-1β and IL-18 in the cells were all significantly increased(all P<0.05);furthermore,the cell prolifer-ation rates,numbers of cell colonies,mean optical density values of GSDMD and cleaved Caspase-1 positive expressions,and the levels of IL-1β and IL-18 in the cells of each herbal decoction+rhNLRP3 group were all increased or decreased with increasing of decoction concentration,and all pairwise comparisons showed statistically significant differences(all P<0.05).Conclusion Prescription for fortifying spleen and dispersing cancer can inhibit the proliferation and promote the py-roptosis of colorectal cancer HCT116 cells through the activation of the NLRP3/caspase-1/GSDMD signaling pathway to re-lease large amounts of IL-1β and IL-18.
