Genetic diversity analysis of Tong sheep rams based on microsatellite markers

[目的]基于微卫星标记分析同羊种公羊的遗传多样性,为同羊养殖业提供科学的家系鉴定与遗传管理方法.[方法]以36只同羊种公羊为对象,提取其血液DNA,使用荧光基团标记的9个微卫星[1个Y染色体微卫星位点(SRYM18)和 8 个常染色体微卫星位点(MAF65、AE129、ILSTS5、JMP29、SRCRSP9、FCB304、MCM140、BM8125)]引物进行PCR扩增,经CERVUS软件完成基因分型分析,利用R 4.4.1软件中的Poppr包计算个体间Nei氏遗传距离,并采用非加权配对算术平均法构建系统聚类图,进而分析同羊种公羊的遗传多样性特征.[结果]除了位于Y染色体的SRYM18外,其余8个位于常染色体的微卫星位点都具有高度多态性;双亲基因型未知时的单亲累计排除概率为 95.27%,单亲基因型已知时的累计排除概率为 99.54%,双亲基因型未知时的双亲累计排除概率为99.99%;36只同羊被划分为9个家系.[结论]对同羊种公羊的家系进行了鉴定与划分,为种公羊的选择和管理提供了科学依据,有利于促进同羊育种工作的可持续发展和遗传改良.

[Objective]This study aims to assess the genetic diversity of breeding rams of the Tong sheep breed using microsatellite markers,in order to provide a scientific basis for pedigree identification and genetic management in Tong sheep farming.[Method]A total of 36 rams of the Tong sheep breed were selected for analysis.Genomic DNA was extracted from blood samples of the rams,and PCR amplification was performed using nine fluorescently labeled microsatellite primers,including one Y-chromosome-specific marker(SRYM18)and eight autosomal markers(MAF65,AE129,ILSTS5,JMP29,SRCRSP9,FCB304,MCM140,BM8125).Genotyping analysis was completed by using CERVUS software.Nei's genetic distances between individuals were calculated by using the Poppr package in R 4.4.1 software,and a systematic cluster diagram was con-structed by using the UPGMA method to analyze the genetic diversity characteristics of Tong sheep rams.[Re-sult]Except for the Y-linked locus SRYM18,all the other eight autosomal microsatellite loci exhibited high polymorphism.The cumulative exclusion probability for single-parent exclusion was 95.27%when both paren-tal genotypes were unknown,the cumulative exclusion probability for single-parent exclusion was 99.54%when one parental genotype was known,and the cumulative exclusion probability for both-parent exclusion was 99.99%when both parental genotypes were unknown.The 36 rams were classified into nine distinct paternal lineages based on UPGMA clustering.[Conclusion]This study successfully identified and categorized the pedi-grees of Tong sheep rams,providing a scientific foundation for ram selection and management,thereby promo-ting the sustainable development and genetic improvement of Tong sheep breeding programs.
