探讨白杨素与木犀草素联合应用对人肾癌细胞 A498 凋亡的影响.收集处于对数生长期的人肾癌细胞A498,实验分为对照组、木犀草素组、白杨素组、二者联合应用组.利用 CCK-8 法测定白杨素、木犀草素和二者联合应用对 A498 细胞生存率的影响.使用激光扫描共聚焦显微镜(CLSM)测定细胞内活性氧(ROS)含量与线粒体膜电位的变化,并用流式细胞仪检测细胞凋亡状况.当木犀草素浓度为 40 μmol/L 时,A498 细胞的生存率为79.28%;白杨素浓度为40 μmol/L 时,A498 细胞生存率为75.32%;40 μmol/L 木犀草素联合40 μmol/L 白杨素时,A498 细胞生存率为74.81%.两者联合用药后,细胞内活性氧含量比对照组高 3.4 倍,线粒体膜电位较对照组明显下降,联合应用后 A498 细胞的凋亡率显著上升.木犀草素和白杨素联合应用对人肾癌细胞 A498 有明显的增殖抑制作用,凋亡率显著增加.

This study aimed to investigate the effect of combined chrysin and luteolin on the apoptosis of human re-nal cell carcinoma cell A498.Human renal cell carcinoma A498 cells in the logarithmic growth phase were harves-ted.The experiment was divided into four groups:control group,luteolin group,chrysin group,and chrysin-luteolin combination group.The CCK-8 assay was used to determine the effects of chrysin,luteolin,and their combination on the survival rate of A498 cells.Laser scanning confocal microscopy(CLSM)was employed to measure intracellu-lar reactive oxygen species(ROS)levels and changes in mitochondrial membrane potential,while flow cytometry was used to detect cell apoptosis.When the concentration of luteolin is 40 μmol/L,the survival rate of A498 cells is 79.28%;when the concentration of poplar extract was 40 μmol/L,the cell survival rate of A498 was 75.32%;when 40 μmol/L luteolin was combined with 40 μmol/L chrysin,the cell A498 survival rate was further reduced to 74.81%.After the combination of the two drugs,the intracellular reactive oxygen species level was 3.4 times higher than the control group,and the mitochondrial membrane potential decreased significantly.The apoptosis rate of A498 cells significantly increased after the combination application.The combined application of luteolin and poplar ex-tract has a significant inhibitory effect on the proliferation and apoptosis rate of human renal cancer cell line A498.
