目的 优化壳聚糖(CS)和海藻酸钠(SA)共修饰结肠靶向和厚朴酚/大黄酸双载药口服脂质体(HNK/RH-CS-SA Lip)的处方工艺,并探讨其结肠靶向性.方法 采用薄膜分散法制备 HNK/RH-CS-SA Lip.以磷胆比、药脂比和超声功率为评价指标,使用单因素考察和BoX-Behnken 响应面法筛选出最佳工艺;对HNK/RH-CS-SA Lip的外观形态、粒径、不同介质中的体外释放度等进行评价;通过小鼠组织药物分布实验,考察HNK/RH-CS-SA Lip体内的结肠靶向性.结果 HNK/RH-CS-SA Lip最佳制备工艺为:磷脂-胆固醇质量比4.44∶1、药物-磷脂质量比1∶9.77,超声功率600 W,水化介质为 PBS,水化体积 6 mL.经响应面优化制得的HNK/RH-CS-SA Lip 平均包封率为92.98%±0.74%,平均粒径为(340.78±2.01)nm,释放行为具有缓释特性,且具有靶向结肠释放特性.结论 制备的HNK/RH-CS-SA稳定好、粒径分布均匀、包封率高,具结肠靶向特性.

Objective To optimize the formulation process of honokiol/rhein dual-loaded oral liposomes modified with chitosan(CS)and sodium alginate(SA)for colon targeting(HNK/RH-CS-SA Lip)and investigate their colon-targeting properties.Methods HNK/RH-CS-SA Lip were prepared using the thin-film dispersion method.With the phospholipid-to-cholesterol ratio,drug-to-lipid ratio,and ultrasonic power as evaluation indices,single-factor experiments and Box-Behnken response surface methodology were employed to screen the optimal preparation conditions.The morphological characteristics,particle size,and in vitro release behavior in different media of HNK/RH-CS-SA Lip were evaluated.The colon-targeting property in vivo was assessed through tissue distribution experiments in mice.Results The optimal preparation conditions for HNK/RH-CS-SA Lip were as follows:phospholipid-to-cholesterol mass ratio of 4.44:1,drug-to-phospholipid mass ratio of 1:9.77,ultrasonic power of 600 W,hydration medium of PBS,and hydration volume of 6 mL.Under the optimized conditions,the average encapsulation efficiency of HNK/RH-CS-SA Lip was 92.98%±0.74%,and the aver-age particle size was(340.78±2.01)nm.The liposomes exhibited sustained-release behavior and colon-targeted release characteris-tics.Conclusion The prepared HNK/RH-CS-SA Lip demonstrate good stability,uniform particle size distribution,high encapsulation efficiency,and colon-targeting properties.
